Potassium bromide is an inorganic salt that dissociates to its composite ions in aqueous solutions at environmental pH and temperature. Comparison of the available data on the various bromide salts have shown that the bromide ion is the relevant ion for determination of the toxicological profile with simple cations such as potassium, sodium or ammonium, that are ubiquitous in nature, having little or no influence on the bromide ion properties. It is therefore justified to read-across data from other inorganic bromide salts to potassium bromide.

Toxicological behaviour: In the rat, bromide half-life is prolonged from approximately 3 days to 25 days on a salt-free diet with tap water.

Applicant's summary and conclusion

Five groups of female Wistar rats (6 rats per group) were all given feed containing 2000 ppm bromide daily ad libitum for 3 weeks. After this the five groups were placed on differing food and drinking water regimes with controlled chloride intake levels. The blood plasma bromide concentrations of all animals were measured 1, 2, 3, 4, 9 and 14 days after completion of the initial 3 weeks of bromide-supplemented feeding.
Chloride depletion prolongs the elimination half-life of bromide. In the rat, bromide half-life is prolonged from approximately 3 days to 25 days on a salt-free diet with tap water. The bromide half-life is shown to be approximately inversely proportional to the total chloride intake. If acceptable daily intakes are to be established, the influence of a salt-free diet on bromide half-life in man will have to be taken into account.
