Test solutions
- Vehicle:
- no
- Details on test solutions:
- - Preparation of stock solution: The test was performed with an aqueous extract (eluate) of the test substance.
- The test substance was stirred in M4 medium for about 20 hours at 20 +/- 2°C.
- Undissolved test substance was removed by centrifugation (approximately 20 min at about 17700 g).
- The nominal concentration of the eluate was 100 mg/L.
Test organisms
- Test organisms (species):
- Daphnia magna
- Details on test organisms:
- - Daphnia magna Straus was originally supplied from "Institut National de Recherche Chimique appliquee, France", and has been breeded from 1978 in the Laboratories of BASF.
- 2 - 24 hours old (starting with the 3rd breed of parent animals)
Study design
- Test type:
- static
- Water media type:
- freshwater
- Limit test:
- no
- Total exposure duration:
- 48 h
Test conditions
- Test temperature:
- 20.3 - 20.9 °C
- pH:
- 7.5 to 8.1
- Dissolved oxygen:
- 4.9 to 8.6 mg/L
- Nominal and measured concentrations:
- The analytical recovery rate was after 0/48 h in all analyzed test solutions below the detection limit (< 1 mg/L). The effective concentrations mentioned here are based therefore on the nominal concentrations of the test substance. Nominal concentrations in mg/L: control, 100, 50, 25, 12.5, 6.25, 3.13, 1.56, 0.78 and 0.39
- Details on test conditions:
- Test conditions:
- Dilution water chemistry: A synthetic fresh water was used for culture and test purposes.
- This water was prepared on the basis of an ultrapure, demineralized water with a conductivity <0.05 uS/cm.
- For the composition of this M4 medium see ISO 10706.
- Properties of this medium:
Total hardness: 2.20 - 3.20 mmol/L
Alkalinity up to pH 4.3: 0.80 - 1.00 mmol/L
Molar ratio Ca:Mg about 4:1
pH value: 7.5 - 8.5
Conductivity: 550 - 650 uS/cm
After preparation the M4 medium is aerated tor approximately 24 h until saturation with oxygen is reached.
- Exposure vessels: test tubes (glass) with flat bottom (nominal volume 20 ml)
- Light conditions: Artificial light, type warm white, day:night rhythm = 16:8 h
- Number of Replicates: 4
- Individuals /replicate: 5
Statistics:
- EC50 values have been calculated with the moving average method.
Measurements:
- pH values and dissolved oxygen: After Oh in an additional replicate without daphnids and after 48h in replicate 1 of each tested concentration.
- Mobile daphnids: Visually after 0, 3, 6, 24 and 48 h
