LC50 (14d) > 1000 mg/L (nominal) for Eisenia fetida (OECD 207)

Additional information
One study investigating the acute toxicity of Bis(tridecyl) adipate (CAS 16958-92-2) to earthworm is available. The study was conducted according to OECD guideline 207 using Eisenia fetida as test organism (Tabor-Kaplon, 2012). Acetone was used as a solvent since the test substance was insoluble in Milli-RO water. Aliquots of acetone stocks were thoroughly mixed with 10 g sand per replicate. Subsequently, after evaporation of the solvent, sand coated with Bis(tridecyl) adipate was mixed with test medium. A limit test was performed exposing forty earthworms (10 per replicate, 4 replicates) to a solvent control and a limit concentration of 1000 mg test substance/kg dw. In addition, ten earthworms (1 replicate) were exposed to 0, 1, 1.0, 10 and 100 mg/kg dw in a combined range-finding test. Bis(tridecyl) adipate did not cause significant mortality of Eisenia fetida at 1000 mg/kg dw after 14 days of exposure (NOEC). The 14-day LC50 was beyond the range tested, i.e. exceeded the limit concentration of 1000 mg/kg dw. According to ECHAs Guidance R.7c, the absence of chronic or long-term effects in aquatic organisms up to the substance solubility limit, can be used as part of a Weight of Evidence argument to modify/waive the data requirements of Annex IX and X. For the category PFAE linear, there are three chronic tests on aquatic invertebrates available for insoluble category members (i.e. CAS 103-23-1 Bis(ethylhexyl)adipate; CAS 33703-08-1 Diisononyl adipate and CAS 103-24-2 Nonanedioic acid, bis(2-ethylhexyl) ester). These substances exhibit log Kow values > 5 and a water solubility < 0.05 mg/L. In none of these tests effects on Daphnia were observed in nominal concentrations well above the limit of water solubility. Therefore, it is assumed that chronic tests on terrestrial organisms will not result in a different outcome. This is supported, by the absence of acute toxicity to earthworm as shown for two other substances within the PFAE linear (i.e. Bis(2-ethylhexyl) adipate CAS 103-23-1 and Dibutyl adipate CAS 105-99-7). Therefore no chronic tests for terrestrial organisms are either provided, or deemed as necessary for the chemical safety assessment.
