Description of key information
The key acute oral study is an Acute Toxic Class study with GLP reporting an LD50 value in the range 25-200 mg/kg bw (Huls AG, 1997). There are no data for the inhalation and dermal routes and the corrosive nature of dichloro(3-chloropropyl)methylsilane means that acute studies via these routes (required in Section 8.5.2 and 8.5.3, Annex VIII) do not need to be conducted. The selected study is the worst-case assessment of acute oral toxicity based on studies with the registered substance. The test was conducted in accordance with an appropriate OECD test guideline and in compliance with GLP.

Two oral Acute Toxic Class studies, with GLP, on dichloro(3-chloropropyl)methylsilane report LD50 values in the ranges 25-200 mg/kg bw (Huls AG, 1997) and 200-2000 mg/kg bw (LPT, 2002). The earlier study giving the lower LD50 value is taken as the key study. It should be noted that in this study all doses are given neat, while in the Wacker study, the lower dose of 200 mg/kg bw is given in corn oil, thus reducing local effects. No data were available for the inhalation or dermal routes.
The supporting acute inhalation studies on trichloro(methyl)silane, trichloro(ethyl)silane and trichloro(propyl)silane are included to support the data waiver for repeated dose toxicity testing via the inhalation route (see Section 5.6.3 of the Chemical Safety Report).
Justification for classification or non-classification
On the basis of the available oral LD50 data, dichloro(3-chloropropyl)methylsilane is classified Acute Toxic 2 (oral) according to Regulation (EC) No 1272/2008.
