Skin irritation

The skin irritating potential of methylpentynol was assessed in an OECD 404 guideline study (BASF, 1990). Three White Vienna rabbits received 0.5 mL undiluted test substance on the shaved skin. The exposure duration was 4 h under semiocclusive conditions (then washing off). The skin reactions were examined 4, 24, 48 & 72 h post treatment. The mean scores of erythema and edema were 0.1 and 0 in three rabbits 24, 48 and 72 h after application. The observed effects were fully reversible within 72 hours. Based on these findings methylpentynol is considered to be non irritating to the rabbit skin.

Eye irritation

In an eye irritation study according to OECD 405 (BASF AG, 1990), 0.1 ml of the undiluted test substance was instilled into the conjunctival sac of one eye of a White Vienna rabbit. The eye was not washed out. The animal was observed for 15 days. After 1 hour, erythema, edema as well as corneal opacity were observed. The mean value of the scores after the 24, 48 and 72 hour reading for erythema, edema, iris and corneal opacity were calculated to be 2.0, 2.0, 0.3, and 1.0, respectively. The observed changes were not reversible within 15 days. Due to the observed effects metylpentynol has the potential to seriously damage the eyes.

Justification for classification or non-classification

According to the criteria of EU Classification, Labelling and Packaging of Substances and Mixtures (CLP) Regulation (EC) No. 1272/2008, classification for eye irritation is warranted (H318).
