- TEST ITEM SOLUTION:
FAT 40'800/A (10.1 mg) was dissolved in methanol (25 mL) to prepare a stock solution of 404 µg/mL. 1 mL of the stock solution was diluted with a solvent mixture of methanol and water (55:45 v/v) to 10 mL to obtain a standard solution of 40.4 µg/mL. The pH value of the test item solution was determined to be 7.2. The test item solution was injected three times.
REFERENCE SOLUTIONS:
The reference items were chosen from the list given in the OECD Guideline No. 121. Stock solutions of the reference items in methanol were prepared as described: 23.7 mg of the reference item phenol, 21.2 mg of isoproturon, 23.7 mg of triadimenol, 22.4 mg of linuron, 23.2 mg of pyrazophos and 24.8 mg of diclofop-methyl were each dissolved in 25 mL methanol.
A combined reference solution was prepared by transferring 220 µL of the phenol, 250 µL of the isoproturon stock solution, 800 µL of the triadimenol stock solution, 140 µL of the linuron, 2 ml of the pyrazophos stock solution and 1 ml of the diclofop-methyl stock solution into one volumetric flask and diluted to a volume of 10 ml with eluent. This reference solution was injected six times.
Sodium nitrate (55 mg) was diluted to 50 mL with methanol to obtain a stock solution. The stock solution (50 pi) was diluted to 1 mL with a solvent mixture of methanol and water (55:45 v/v). The sodium nitrate solution was injected two times for the determination of the dead time of the HPLC system.

This method utilises a high performance liquid Chromatograph with UV-VIS detector, reversed phase column and an aqueous mobile phase. Details:
