The test substance is covered by the category approach of methylenediphenyl diisocyanates (MDI). Hence, data of the category substances can be used to cover this endpoint. The read-across category justification document is attached in IUCLID section 13. It is important to note that the MDI category approach for read-across of environmental and human hazards between the MDI substances belonging to the MDI category is work in progress under REACH. Therefore the read-across document should be considered a draft.
Most aquatic tests were not performed with the test substance, but with an analogue covered by the category approach of the methylenediphenyl diisocyanates (MDI).
The key fish study was done with Danio rerio according to OECD 203 guideline (static) (Caspers, 1986. No mortality was observed at 1000 mg/L. No long-term toxicity study in fish is available. However, there is no need to perform a chronic fish study due to the high reactivity of the substance with water.
One short-term study with Daphnia magna is available for polymeric MDI (CAS 9016-87-9). The 24h -EC50 is >1000 mg/L in an OECD 202 test (Caspers et al., 1986). One long-term study with D. magna is available for polymeric MDI (CAS 9016-87-9). The 21d-NOEC is >10 mg/L in an OECD 211 test (Caspers et al., 1986). Although a chronic Daphnia value is determined, this value is not taken into account for PNEC derivation due to the high reactivity of the substance with water.
One freshwater study with the green alga Scenedesmus subspicatus is available for polymeric MDI (CAS 9016-87-9). The 72h-EC50 and 72h-NOEC is 1640 mg/L based on growth rate and nominal concentrations. The test was conducted under static conditions and according to OECD 201 (Blom and Oldersma, 1994).
One OECD 209 Activated Sludge, Respiration Inhibition Test is available for polymeric MDI (CAS 9016-87-9). Based on the respiration rate using activated sludge, the 3h-EC50 is found to be >100 mg/L (Caspers et al. 1986).
These values may be considered for the derivation of a PNECaqua and a PNECstp.
Based on the justification in the category approach of methylenediphenyl diisocyanates (MDI), it is assumed that all MDI analogues included in the category have similar toxicity effect concentrations.
All aquatic toxicity studies have been conducted in freshwater. However, the hydrolytic behaviour of MDI in freshwater and seawater is very similar (PMDI, Yakabe et al, 1994). Therefore, the toxicity of MDI substances of the category approach and their hydrolytic degradation products to both freshwater and the corresponding marine water organisms is expected to be similar.
