Applicant's summary and conclusion

Executive summary:

MCPP in range-finding experiment in prokaryotic modelSalmonella typhimuriumwas tested up to a maximum dose 5 mg/plate; this dose selected according OECD 471 guideline as the highest tested dose was not toxic.MCPP dissolved in dimethylsulphoxide was evaluated for mutagenicity in Ames standard plate assay in concentration range of 0.039 - 1.25 mg/plate in experiments both in the presence and absence of S9 Mix. The higher concentrations of MCPP reduced the number of spontaneous revertants. In confirmation tests the standard plate test was modified with liquid preincubation and with use of other S9 preparation.

No substantial increases in the number of revertant colonies were observed in any of the fiveSalmonella typhimuriumstrains. Adequate positive and negative controls were performed and showed the reliability of the test system. The MCPP did not produce either a statistically significant dose-related increase in the number of revertants nor a statistically significant and reproducible positive response at any one of the test points and according these results is considered nonmutagenic in prokaryotic system for detection of gene mutations.
