Medium 1 : Simulated saliva : Dissolve 4.2 g of sodium bicarbonate (NaHCO3), 0.5 g of sodium chloride (NaCl), and 0.2 g of potassium carbonate (K2CO3) in 1 litre of distilled water or water of equivalent quality. The solution should be approximately pH 9.

Medium 2 : Simulated gastric fluid ; HCl 0.07 M (pH 1.15)

Medium 3 : Simulated intestinal fluid : Dissolve 6.8 g of KH2PO4 in 250 ml of water and add 190 ml of 0.2 M NaOH and 400 ml of water. Add 10.0 g of pancreatin, mix, and adjust the resulting solution with 0.2 M NaOH to a pH of 7.5-+ 0.1 . Dilute with water to 1000 ml.

The potential of the test substance to hydrolyse under environmental conditions was investigated in a hydrolysis test in 3 different physiological media at pH 1.15, 7.5 and 9. SPM has been dissolved in media 1 (Simulated saliva, pH 9), 2 (Simulated gastric fluid, pH 1.15) and 3 (Simulated intestinal fluid, pH 7.5) in a concentration of approx. 0.1 w/v-%. The hydrolysis has been measured after 0, 1, 2 and 4 hours at 37°C. SPM and the hydrolysis product Methacrylic acid have been determined by HPLC. 0 - 16 ppm of SPM led to approx. 0 - 6 ppm (theoretical)/0 - 5 ppm (actual) Methacrylic acid and to approx. 0 - 12 ppm (theoretical) Potassium-3-hydroxypropane-1-sulfonate.
