PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: Guidance document on aquatic toxicity testing of difficult substances and mixtures, OECD series on testing and assessment number 23, 2000

Preparation of test solutions started with a loading rate of 100 mg/L applying a one day period of magnetic stirring to ensure maximum dissolution of the test item in medium. The obtained mixture was allowed to settle for 13 minutes. Thereafter, the aqueous Saturated Solution (SS) was collected by means of siphoning and used as the highest test concentration. Lower test concentrations were prepared by subsequent dilutions of the SS in test medium. All test solutions were clear and colorless at the end of the preparation procedure.

Validity criteria fulfilled:
yes
Conclusions:
In conclusion, the 48h-EC50 for Daphnia magna exposed to Tri-n-butyl phosphine equalled a concentration representing 9.2% of a saturated solution prepared at a loading rate of
100 mg/L (95% confidence interval between 7.5% and 11% SS).
Executive summary:

The acute toxicity of the test material to freshwater invertebrates was determined in a study conducted in accordance with the standardised guideline OECD 202 under GLP conditions. Daphnia magna were exposed to the test material for 48 hours under static conditions at nominal concentrations 0.46%, 1.0%, 2.2%, 4.6%, 10% and 22% of a SS prepared at a loading rate of 100 mg/L (no solvent). A SS prepared with loading rate was used due to the poor solubility of the test substance.

Under the conditions of the test, the 48h EC50 of the test material to Daphnia magna was determined to be 9.2% of a SS prepared at a loading rate of 100 mg/L.
