Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
- Concentrations: the test substance was dissolved directly in water at 500 mg/l abd agitated mechanically for around 24 hours in an ttempt to completely dissolve it. Test solutions were prepared by further dilution of the stock solution with water to provide a geometric series of concentrations: 0, 1, 10 and 100 mg/l

- Sampling method and sample storage conditions before analysis: a 4 ml sample was taken from the limit test solution and the dilution water at the beginning of the limit test and frozen, and a further 4 ml sample was taken from the limit vessel containing fish and the control vessel at the end of the test. A sample of 4 ml was taken on 5 occasions during the test (T0, T24, T48, T72, T96) from the 10 mg/l (nominal) test solution and was analysed to verify the concentrations of test substancz throughout the test.

Test organisms

Oncorhynchus mykiss (previous name: Salmo gairdneri)
Details on test organisms:
TEST ORGANISM
- Common name: rainbow trout
- Source: "Labbas", Le Hamet, F-27400 Acquigny (France)
- Length at study initiation (length definition, mean, range and SD): average: 46.4 mm, ranging from 40 to 56 mm (based on a measurement of a sample of 30 fishes used in the test)
- Weight at study initiation (mean and range, SD): the mean weight based on a sample of 10 fishes removed at random from the acclimatation tank was 1.7 g
- Feeding during test: no

ACCLIMATION
- Acclimation period: animals were acclimated for 7 days in a flow-through system in water of the same quality and temperature as that to be used in the test.
- Type and amount of food, feeding frequency: during acclimatation, animals were fed trout chow twice daily, ceasing 24 hours before the start of the test.

Applicant's summary and conclusion

Validity criteria fulfilled:
not specified
Conclusions:
The test substance 852 DIESTER is toxic to Oncorhynchus mykiss in a static test at the nominal concentration of 100 mg/l but non toxic at the measured concentration of 7.23 mg/l.
Although the maximum concentration which did not result in mortality was measured as 7.23 mg/l (geometric mean), it is considered unlikemy that serious toxic effects would have been observed at 10 mg/l had it been possible to maintain this concentration.
The test substance 852 DIESTER should not be classified.
