fish short-term toxicity test on embryo and sac-fry stages
Type of information:
other: published data
Adequacy of study:
weight of evidence
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
study well documented, meets generally accepted scientific principles, acceptable for assessment
Justification for type of information:
Carbon disulphide is both a reagent in the manufacture, as well as a decomposition product of xanthates. SIBX readily decomposes to carbon disulphide, especially in the presence of moisture/water. Therefore, the health effects of carbon disulphide (CS2) need to be considered in the assessment of SIBX.
Qualifier:
equivalent or similar to guideline
Guideline:
OECD Guideline 212 (Fish, Short-term Toxicity Test on Embryo and Sac-Fry Stages)
Deviations:
no
Remarks:
The test was carried out for maximally 10 days in closed vessels, with renewal of test solution every 2-3 days. The actual concentration was not measured. The test did not follow a particular guideline (not available at the time the study was performed),
Principles of method if other than guideline:
Eggs from Danio rerio were introduced in the test solutions. Incubation started within 4 hours after spawning. Daily observations were made on survival of eggs and larvae, hatching and malformations. The test was semi-static with renewal after 2 - 3 days in closed containers.

Specific malformations were observed in the development of the notochord 2 days after fertilisation. The notochords were abnormally stretched and were twisted and distorted. Malformations in the muscle tissue and organ dislocations were also observed. Other sublethal effects: oedemous bodies, failure to hatch or to shed the eggshell, lumpy skin, incomplete fins.

Conclusions:
The Sodium isobutyl xanthate (CAS No. 25306-75-6), the subject of this dossier) is expected to exhibit very similar toxicity to Carbon disulphide (CS2, (CAS No. 75-15-0)), the decomposition product of Sodium isobutyl xanthate. Comparable metabolism would occur.
On a molecular weight scaled basis, the NOEC would be 5.66mg/l.
(2.5 x 172.24) /76.13 =5.66 mg/l
Executive summary:

The effect of CS2 was studied in an Early Life Stage test with Danio rerio (zebra fish). The test resembled the OECD 212 test, but was carried out in sealed vessels for 8 days to prevent the escape of volatile CS2.
