96 -Hour Acute Toxicity Study in Carp with Benzenesulfonic acid, 4-C15-16-sec-alkyl derivs..
The study procedure described is based on the OECD guideline No. 203, 1992. In addition, the procedures were designed to meet the test methods of the Comission Regulation (EC) No 440/2008, Part C.1, 2008 and the ISO International Standard 7346 -1: Static method, 1996.
The batch of Benzenesulfonic acid, 4-C15-16-sec-alkyl derivs. tested was a dark brown viscous liquid with a purity of 89.2%. Stock solutions were prepared in acetone at concentrations a factor of 10,000 higher than the target concentrations.
A final test under flow-through conditions was performed based on the results of a preceding range-finding test under static conditions. The stock solutions were dosed via a computer-controlled system from the dilution water (ISO-medium) supply. The dilution water was supplied via a flow meter with a flow rate of 6 L/h. In the mixing flasks the dosed volumes and the dilution water were mixed under continuous stirring. Subsequently, the test solution coming from the mixing flask entered the vessel containing the fish. The whole system was checked daily.
Seven fish per concentration were exposed to a control and to target concentrations of 0.046, 0.10, 0.22, 0.46 and 1.0 mg/L. The total exposure period was 96 hours and samples for analytical confirmation of actual exposure concentrations were taken two days before the start of exposure, at the start, after 48 hours of exposure and at the end of the test (from the control and all concentrations with surviving fish). In addition samples were taken after 24 hours of exposure from the two highest test concentrations.
Analysis of the samples taken two days prior to the start of the test showed that the system was functioning adequately although most recoveries were somewhat low relative to the target concentrations, i.e. 62 -84% of target, with one exception for the lowest test concentration at the start of the test (105% of target). Given these results, the effect parameters were based on the average exposure concentrations that were calculated to be: 0.039, 0.075, 0.16, 0.34, and 0.81 mg/L.
The study met the acceptability criteria prescribed by the protocol and was considered valid.
The 96h-LC50 was 0.17 mg/L (95% confidence interval between 0.13 and 0.24 mg/L) and was already reached after 72 hours of exposure.

The obtained 96h-LC50 of 0.17 mg/L (95% CL 0.13 -0.24 mg/L) in fish is based on the average exposure concentrations. Acetone was used as solvent to enable stable concentration in the flow through system. This is not considered to have any impact on the result of the study, as the acute toxicity value (48h-EC50) for fresh water invertebrates lies in the same range.
