Details on study design: HPLC method:
For the estimation of the adsorption coefficient the HPLC-method was used. Therefore, an analytical column packed with a commercially available, cyanopropyl solid phase containing lipophilic and polar moieties was used. A moderately polar stationary phase based on a silica matrix was used. While passing through the column along with the mobile phase the test item interacts with the stationary phase. As a result of partitioning between mobile and stationary phases the test item is retarded. The dual composition of the stationary phase having polar and non-polar sites allows for interaction of polar and non-polar groups of a molecule in a similar way as it is the case for organic matter in soil or sewage sludge matrices. This enables the relationship between the retention time on the column and the adsorption coefficient on organic matter to be established.

Details on stock solution:
31.24 mg of FAT 40224/C were dissolved in 20 mL of water to prepare a stock solution of 1562.0 µg/mL FAT 40'224/C. 1 mL of the stock solution were diluted with a solvent mixture of methanol and water (45:55 v/v) to 20 mL to obtain a standard solution of 78.1 µg/mL FAT 40224/C.

The determination of the adsorption coefficient (Koc) of FAT 40224/C was performed according to the OECD Guideline for the Testing of Chemicals, Estimation of the Adsorption Coefficient (Koc) on Soil and Sewage Sludge using High Performance Liquid Chromatography (HPLC), Draft Guideline (June 2000).

The test item solution was injected three times and the reference item solutions were injected six times. The retention time of the test item solution was shorter than the retention time of the lowest retention time of the reference items (Koc = 21, log Koc= 1.32). This indicated that FAT 40224 is mainly dissociated. The log Koc was calculated using a regression curve (log k' vs. log Pow) and was found to be <1.32.

Based on the chromatographic data, FAT 40224/C was considered to be stable during the test procedure. Using the corresponding log Koc values of the reference items from the OECD guideline, the regression coefficients were calculated. The log Koc value of the test item was calculated from the calibration equation. In conclusion, the adsorption coefficient (K0c) of FAT 40224/C was estimated using the HPLC method to be Koc = <21. This value indicates a mobile character of FAT 40224/C.
