- Les animaux ont recu chaque concentration de la substance d'essai sur le dos des deux oreilles quotidiennement pendant 3 journées.
Les rats ont reçu 25 µl de substance ou de véhicules. Les animaux ont été sacrifiés 24 h après l'exposition finale. Les ganglions lymphatiques auriculaires drainants ont été excisés et regroupés et pesés pour chaque groupe expérimental. Une seule suspension de LNC était préparé par désagrégation mécanique à travers un acier inoxydable stérile. Les suspensions cellulaires ont été lavées une fois dans une solution saline tamponnée au phosphate (pH 7,2) et resuspendue dans du milieu de culture RPMI-1640 additionné de 10% de SVF, 25 mM Acide N-2-hydroxyéthylpipérazine-N-2-éthanesulfonique (HEPES), 100 tg / ml de pénicilline, 100 unités / ml de streptomycine (FCS-RPMI). Le nombre total de LNC a été déterminé en utilisant un compteur de cellules automatisé. Des suspensions cellulaires (1 x 106 cellules / 200 / ml) ont été ensemencées dans des plaque de culture tissulaire à 96 puits (5 puits par groupe) et cultivée à 37 ° C dans un milieu humidifié, atmosphère de 5% de CO2 dans l'air avec 0,5 / zCi [méthyl-3H] thymidine (3HTdR). Après 18h culture, LNC ont été récoltés avec une abatteuse de cellules automatique et incorporation 3HTdRa été déterminée par comptage par scintillation liquide - Positive control substance(s):
