Short-term toxicity to fish
A study was predicted using OECD QSAR toolbox version 3.3 (2017) with respect to the descriptor log Kow; to assess the toxicity effect of disodium 2-[[5-carb amoyl-1-ethyl-1,6-dihydro-2-hydroxy-4-methyl-6-oxo-3-pyridyl] azo]-4-[[4-chloro-6-[[3-[[2-(sulphonatooxy)ethyl]sulphonyl]phenyl] amino]-1,3,5-triazin-2-yl] amino] enzenesulphonate on fish in a 96 hours of exposure. The median lethal concentration (LC50) for the test substance, disodium 2-[[5-carbamoyl-1-ethyl-1, 6-dihydro-2-hydroxy-4-methyl-6-oxo-3-pyridyl] azo]-4-[[4-chloro-6-[[3-[[2-(sulphonatooxy) ethyl]sulphonyl]phenyl]amino]-1,3,5-triazin-2-yl]amino] benzenesulphon ate, in Danio rerio on the basis of mortality effect was estimated to be 484.78 mg/L. Thus, on the basis of this LC50 value and according to CLP Criteria for aquatic classification of the substance, it is concluded that disodium 2-[[5-carbamoyl-1-ethyl-1,6-dihydro-2-hydroxy-4-methyl-6-oxo-3-pyridyl] azo]-4-[[4-chloro-6- [[3-[[2- (sulphonatooxy)ethyl]sulphonyl]phenyl]amino]-1,3,5-triazin-2-yl]amino]benzenesulphonate (CAS No. 84000-63-5) does not exhibit short term toxicity to fish.
Short-term toxicity to aquatic invertebrates
Using the OECD QSAR toolbox version 3.3 with log kow as the primary descriptor and considering the six closest read across substances, the short term toxicity on aquatic invertebrate was predicted for disodium 2-[(5-carbamoyl-1-ethyl-2-hydroxy-4-methyl-6-oxo-1,6-dihydropyridin-3-yl)diazenyl]-4-({4-chloro-6-[(3-{[2-(sulfonatooxy)ethyl] sulfonyl}phenyl)amino]-1,3,5-triazin-2-yl}amino)benzenesulfonate (Cas no. 84000-63-5). Effect concentration i.e EC50 value was estimated to be 541.0 mg/l for Daphnia magna for 48 hrs duration. Based on the effect value, the substance was likely to be non toxic to aquatic invertebrates at environmentally relevant concentrations and hence it can be considered to be not classified as per the CLP classification criteria.
Toxicity to aquatic algae and cyanobacteria
Using the OECD QSAR toolbox version 3.3 with log kow as the primary descriptor and considering the five closest read across substances, the short term toxicity on aquatic algae and cyanobacteria was predicted for target substance disodium 2-[(5-carbamoyl-1-ethyl-2-hydroxy-4-methyl-6-oxo-1,6-dihydropyridin-3-yl)diazenyl]-4-({4-chloro-6-[(3-{[2-(sulfonatooxy)ethyl] sulfonyl}phenyl)amino]-1,3,5-triazin-2-yl}amino)benzenesulfonate (CAS no. 84000-63-5). EC50 value was estimated to be 376.64 mg/l for Pseudokirchneriella subcapitata for 72 h duration. Based on this value it can be concluded that the substance is likely to be non toxic at environmentally relevant concentrations and can be considered to be not classified as per the CLP classification criteria.
Toxicity to microorganisms
A study was predicted using OECD QSAR toolbox version 3.3 with respect to the descriptor Primary biodeg (Biowin 4), to assess the toxicity effect of disodium 2-[[5-carbamoyl-1-ethyl-1 ,6-dihydro-2- hydro xy-4-methyl-6-oxo-3-pyridyl] azo]-4-[[4-chloro-6-[[3-[[2-(sulphonatooxy)ethyl]sulphonyl]phen yl]amino]-1,3,5- triazin- 2-yl]amino]benzene sulphonate (CAS No.84000-63-5) in microorganism (tetrahymena pyriformis) in a 48 hours of exposure. The inhibition growth concentration (IGC50) of disodium 2-[[5-carbamoyl-1-ethyl-1,6-dihydro-2-hydroxy-4-methyl-6-oxo-3-pyridyl] azo]-4-[[4- chloro-6- [[3-[[2-(sulphonatooxy)ethyl]sulphonyl]phenyl] amino] -1,3, 5-triazin-2-yl]amino]benzenesulphonate in microorganism (Tetrahymena pyriformis) on the basis of growth inhibition effect was estimated to be 27.809 mg/L.

Moreover, the fish acute toxicity test according to OECD Guideline 203 was conducted by SSS (Europe) AB (2013) for read across substance disodium 6- hydr oxy-5-[(2-methoxy-4-sulphonato-m-tolyl) azo] naphthalene -2- sulphonate (ALLURA RED AC) (CAS No.- 25956-17-6) on Zebra fish(Danio rerio) in a 96 hour study. The nominal concentration selected for the experiment were 100 mg/L and test fish were exposed to these concentration for 96 hours. The median lethal concentration (LC50) for disodium 6-hydroxy-5-[(2-methoxy- 4-sulphonato-m-tolyl) azo] naphthalene-2-sulphonate on Danio rerio in a 96 hours study on the basis of mortality effect was found to be >100 mg/L.
Based on the above predicted and experimental studies for target substancedisodium 2-[[5-carbamoyl-1-ethyl-1,6-dihydro-2-hydroxy-4-methyl-6-oxo-3-pyridyl] azo]-4-[[4-chloro-6-[[3-[[2-(sulphonatooxy)ethyl] sulphonyl]phenyl]amino]-1,3,5-triazin-2-yl]amino]benzenesulphonate(CAS No. 84000-63-5) and to its read across substance ALLURA RED AC (CAS No.- 25956-17-6), the lethal concentration LC50 value was found to be in the range of >100.0 mg/L to 484.78 mg/L. Thus, on the basis of this LC50 value and according to CLP Criteria for aquatic classification of the substance, it is concluded thatdisodium 2-[[5-carbamoyl- 1-ethyl-1,6-dihydro-2-hydroxy-4-methyl-6-oxo-3-pyridyl] azo]-4-[[4-chloro-6-[[3-[[2-(sulphonatooxy)ethyl]sulphonyl]phenyl] amino]-1,3,5-triazin-2-yl] amino]benzene sulphonate(CAS No. 84000-63-5) does not exhibit short term toxicity to fish.

First data from prediction using the OECD QSAR toolbox version 3.3 with log kow as the primary descriptor and considering the six closest read across substances, wherein the short term toxicity on aquatic invertebrates was estimated for disodium 2-[(5-carbamoyl-1-ethyl-2-hydroxy-4-methyl-6-oxo-1,6-dihydropyridin-3-yl)diazenyl]-4-({4-chloro-6-[(3-{[2-(sulfonatooxy)ethyl] (Cas no. 84000-63-5). The median Effect concentration i.e EC50 value was estimated to be 541.0 mg/l for Daphnia magna for 48 hrs duration.
Above predicted result of target assist by experimental result of read across chemical Dimethylformamide (Cas no. 68-12-2) J-CHECK authoritative database which indicate Short term toxicity to Daphnia magna (Invertebrates) study was carried out for 48 hrs. according to OECD Guideline 202 (Daphnia sp. Acute Immobilisation Test).The study was based on the effects of the test compound Dimethylformamide (Cas no. 68-12-2) on Daphnia magna in a semi-static fresh water system. Based on Immobilisation effects the 48 hr EC50 value was observed to be > 1000 mg/l.
And from ECOTOX database for another read across 2,2',2'',2'''-(ethane-1,2-diyldinitrilo)tetraacetic acid (Cas no. 60-00-4) indicate Short term toxicity to aquatic invertebrates was performed in Daphnia magna (Water Flea) for 48 hrs with <24 h age (Neonate).Test in fresh water media with static condition and at 20 deg.C test temp. During the experiment, the EC50 value for 2,2',2'',2'''-(ethane-1,2-diyldinitrilo)tetraacetic acid (Cas no. 60-00-4)was determined to be 113 mg/l on the basis of mobility.
Same data source (ECOTOX database) for read across chemical 2-[({[(4,6-dimethoxypyrimidin-2-yl)carbamoyl]amino}sulfonyl)amino]-N,N-dimethylbenzamide (Cas no. 213464-77-8) indicate Short term toxicity to aquatic invertebrates was performed in Americamysis bahia (Opossum Shrimp) for 96 hrs with <24 h age (Neonate).Test in salt water media with static condition. During the experiment, the LC50 value for 2-[({[(4,6-dimethoxypyrimidin-2-yl)carbamoyl]amino}sulfonyl)amino]-N,N-dimethylbenzamide (Cas no. 213464-77-8)was determined to be 122 mg/l on the basis of mortality.
Thus based on the all available effect concentrations which is in the range of 113 mg/l to 1000 mg/l and lethal concentration (LC50) 122 mg/l for target and read across chemicals give the conclusion that test substance disodium 2-[(5-carbamoyl-1-ethyl-2-hydroxy-4-methyl-6-oxo-1,6-dihydropyridin-3-yl)diazenyl]-4-({4-chloro-6-[(3-{[2-(sulfonatooxy)ethyl] (Cas no. 84000-63-5) was likely to be non toxic to aquatic invertebrates at environmentally relevant concentrations and hence it can be considered to be not classified as per the CLP classification criteria.

Thus based on the effects from both studies give the conclusion that test substance disodium 2-[(5-carbamoyl-1-ethyl-2-hydroxy-4-methyl-6-oxo-1,6-dihydropyridin-3-yl)diazenyl]-4-({4-chloro-6-[(3-{[2-(sulfonatooxy)ethyl] (Cas no. 84000-63-5) was likely to be non toxic to aquatic algae at environmentally relevant concentrations and can be considered to be not classified as per the CLP classification criteria.

Moreover, a test was developed by Sako,F. et al. (Toxicol. Appl. Pharmacol.39:111-117,1977)using Paramecium caudatum (Ciliate) in order to determine the toxicity of disodium 6-hydroxy-5-[(4-sulphonatophenyl)azo]naphthalene -2-sulphonate (CAS No.- 2783-94-0). Paramecium caudatum was exposed for 0.012 days. The concentration of the chemical, at which time to 50% mortality of Paramecium caudatum observed (LT50), was determined. The median lethal time (LT50) of disodium 6-hydroxy-5-[(4-sulphonatophenyl)azo] naphthalene-2- sulphonate (CAS No.- 2783-94-0) in microorganism (Paramecium caudatum (Ciliate)) in a 0.012 days study on the basis of mortality effect was observed to be 1000.0 mg/L.
Thus, based on the above predicted and experimental studies for target substancedisodium 2-[[5-carbamoyl-1-ethyl-1,6-dihydro-2-hydroxy-4-methyl-6-oxo-3-pyridyl] azo]-4-[[4-chloro-6-[[3-[[2-(sulphonatooxy)ethyl]sulphonyl]phenyl]amino] -1,3,5-triazin- 2-yl]amino]benzenesulphonate(CAS No. 84000-63-5) and to its read across substance disodium 6-hydroxy-5-[(4-sulphonatophenyl)azo] naphthalene-2- sulp honate (CAS No.- 2783-94-0), the inhibition growth concentration (IGC50) value was found to be 27.809 mg/L and LT50 value to be 1000.0 mg/L on the basis of various effects (growth inhibition and mortality effect) in a 0.012 days to 48 hours of exposure.
