- Seven replicates of test item at 20 mg/L loading rate were inoculated at 5 017 cells per mL. A eight replicate non inoculated was used as colour control. A control (6 replicates) was tested under the same conditions with test media only.

- the test item solution was prepared by dissolution of 20.0 mg of butyl avocadate distillated in 1 000 mL of the test medium to obtain a nominal concentration at 20 mg/L (experimental concentration = 20 mg/L). The solution was prepared in 2 L glass bottle and shaken for about 24h. Then this preparation was filtered through 0.2 µm filter (Whatman cellulose nitrate membrane filters). This solution was used for exposure and identified as saturation concentration.
The pH of this resulting solution was 7.8.

- The test was conducted in 250 mL glass flasks containing 100 mL of test solution under agitation in a climatic chamber. Flasks were incubated at a temperature of between 23.0 to 24.1°C with continious light in a range of 5 600 lux.
After 2', 48 and 72 hours, optical density (683 nm) were measured to determine cell concentrations in each replicate.
At the beginning of the test, test item solution was sampled (volume: 100 mL). After 24, 48 and 72 hours, test item solution was sampled (volume: 100 mL) in the seventh inoculated replicate of saturation concentration solution. These samples were quantified immediately after sampling according to the validated method.

Objective
The scope of this study was, in compliance with the principles of Good Laboratory Practice (GLP), to determine the toxicity of butyl avocadate distillated on Pseudokirchneriella subcapitata (Algae) in a 72 -hour static test.
Method Summary
Saturation concentration in test medium was prepared and divided into 8 replicates. 7 replicates were inoculated in the range of 5 000 to 10 000 cells per mL. The 7th replicate was used as a colour control (not inoculated). 6 inoculated replicates were used to measure algal growth and 1 inoculated replicate was used for analytical monitoring.
A control (6 replicates) was tested at the same time with test medium inoculated in the range of 5 000 to 10 000 cells per mL.
