Description of key information

The toxicity of the test itemto the unicellular freshwater green algaPseudokirchneriella subcapitatawas determined according to the principles of OECD 201 andCouncil Regulation (EC) No. 761/2009/Method C.3.The aim of the study was to assess the effects on growth rate and yield over a period of 72 hours. The study was conducted under static conditions with an initial cell density of 6305 cells/mL. Five water accommodated fractions (WAF) were prepared in a geometrical series with a factor of 2: 12.5 – 25.0 – 50.0 – 100 – 200 mg/L.For each loading level, an appropriate amount of the test item was weighed out. The test item was applied onto a plastic slide. The plastic slide with the test item was inserted in a PE-bottle with an appropriate amount of dilution water. These dispersions were shaken for at least 24 hours with 20 rpm at room temperature. After a separation phase of 1 hour, the WAF were removed by siphoning an used in the test. Three replicates were tested for each test item loading and six replicates for the control. The environmental conditionswere within the acceptable limits. The test solutions were clear and the test item was dissolved throughout the test period. The concentrations of the test item were analytically verified via analysis of total organic carbon (TOC, according to DIN EN 1484) at the start of the exposure (fresh media, 0 hours) and at the end of the exposure (old media, 72 hours). The nominal EL50-values for inhibition of growth rate (ErL50) and yield (EyL50) after 72 hours were > 200 mg/L and 54.2 (27.6 – > 200). The nominal NOEL-values for inhibition of growth rate and yield after 72 hours were for both < 12.5 mg/L, respectively. No clear concentration response relationship was observed. Since water accommodated fractions were tested, different compounds can be responsible for a slight inhibition that was observed in all concentration levels. With one exception, the inhibition of the growth rate was below 10%. However, the inhibition was statistically significant.

Key value for chemical safety assessment

EC50 for freshwater algae:
200 mg/L
EC10 or NOEC for freshwater algae:
53.9 mg/L
