Experimental data on the chronic toxicity of 1,2-Ethanediamine, N-{3-(trimethoxysilyl)propyl}-,N-{(ethenylphenyl)methyl}derivate,hydrochlorides (CAS 171869-89-9) towards freshwater fish are not available. Therefore, in accordance with the Regulation (EC) No. 1907/2006 Annex XI, 1.5 “Grouping of substances and read across” and the read across assessment framework (RAAF, ECHA 2017) a read across from the analogue substance reaction mass containing six benzylated amine silanes (EC 700-961-2) has been performed. Details on the read-across justification can be found in the attached justification in the respective target entry and in the overall justification for grouping of substances attached in IUCLID Section 13.

The study (2004) testing the long term toxicity of reaction mass containing six benzylated amine silanes (EC 700-961-2) to fish was conducted according to OECD guidance 215 using Oncorhynchus mykiss as the test organism. The test item degrades fast in water into oligomeric and polymeric silicon compounds and methanol. According to the OECD Guidance Document on Aquatic Toxicity Testing of Difficult Substances and Mixtures, substances which hydrolyse and form polymers, a flow through exposure system is not considered appropriate. Therefore, for the long term test a semi-static test design with a test medium renewal of three times per week was chosen. Since reaction mass containing six benzylated amine silanes (EC 700-961-2)reacts immediately with water, the biological results are based on the nominal concentrations of the test item. In the 28 day juvenile growth test, rainbow trout was exposed to nominal test item concentrations of 0.5, 1.6, 5, 16 and 50 mg/L. The highest concentration tested without toxic effects (NOEC) was determined to be 5.0 mg/L (nominal). The lowest concentration tested with toxic effects (LOEC) amounted 16 mg/L due to the statistically significantly reduced growth of the test fish at this test concentration. It is assumed that the test organisms were primarily exposed to the hydrolysis products of the substance.

Based on the results of the analogue substance and as justified in the attached justification in the target IUCLID entry, similar results are expected from the target substance 1,2-Ethanediamine, N-{3-(trimethoxysilyl)propyl}-, N-{(ethenylphenyl)methyl}derivate, hydrochlorides (CAS 171869-89-9).
