Rationale for reliability incl. deficiencies:
test procedure in accordance with national standard methods
Qualifier:
according to guideline
Guideline:
other: Norme Experimentale AFNOR T 90-301.
Deviations:
yes
Remarks:
TK 12 184 is not water soluble; stable suspensions were used
GLP compliance:
not specified
Specific details on test material used for the study:
TEST MATERIAL (as stated in study report): TK 12 184; trade name D 16-51

SOURCE OF TEST MATERIAL
- Source and lot/batch No.of test material: Sponsor / Batch No. Mixt. 4/5/6

STABILITY AND STORAGE CONDITIONS OF TEST MATERIAL
- Storage condition of test material: room temperature (20 - 24 deg C)
Analytical monitoring:
no
Vehicle:
no
Details on test solutions:
TK 12 184 was not sufficiently water soluble for the preparation of a 100 mg/L aqueous stock solution. A Suspension of 100 mg/L TK 12 184 in water with an acceptable stability was obtained applying 400 W ultrasound during 4 minutes.
Test organisms (species):
Daphnia magna
Details on test organisms:

METHOD FOR PREPARATION AND COLLECTION OF EARLY INSTARS OR OTHER LIFE STAGES: Animals retained on the 560 pm mesh sieve (sieved 24 h before the test and the day of test)
Test type:
static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
24 h
pH:
8.0
Dissolved oxygen:
O2 = 8.45 mg/L (99 % of Saturation) 22 deg C. There is no valid explanation for the observed oversaturation of oxygen at the end of the test.

Dose descriptor:
IC50
Effect conc.:
12.54 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mobility
Remarks on result:
other: Corrected with the bichromate value
Results with reference substance (positive control):
The IC50 (24 h) of the Bichromate was calculated as 1.1 mg/L.
Conclusions:
Under these test conditions, the IC50 (24 h) for TK 12 184 in Daphna magna, corrected with the Bichromate-value, was calculated as 12.54 mg/L.
Endpoint:
short-term toxicity to aquatic invertebrates
Type of information:
(Q)SAR
Adequacy of study:
supporting study
Study period:
2018
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
results derived from a valid (Q)SAR model and falling into its applicability domain, with adequate and reliable documentation / justification
Principles of method if other than guideline:
Results predicted by US EPA ECOSAR v1.11 model.
GLP compliance:
no
Details on results:
No effects are expected at saturated concentrations in water. The estimated logKow for this substance is greater than the class specific cut-offs for acute Daphnid toxicity, and the predicted effect level exceeds the water solubility of this substance by more than 10x.
Validity criteria fulfilled:
not applicable
Conclusions:
The US EPA ECOSAR v1.11 model predicts no effects at saturated concentrations in water for TGEPO (CAS No. 83547-95-9). The estimated logKow for this substance is greater than the class specific cut-offs for acute Daphnid toxicity, and the predicted effect level exceeds the estimated water solubility of this substance by more than 10x.

Description of key information

The IC50 (24 h) for water suspensions of TK 12 184 in Daphna magna, corrected with the Bichromate-value, was calculated as 12.54 mg/L. Water suspensions were prepared with the aid of high speed mixing due to the very low water solubility of the test substance.

Key value for chemical safety assessment
