Reconstituted water (so-called 'M4 medium', originally described in Water Research 24 (9): 1157-1167), prepared according to the recommendations of Bundesgesundheitsamt Berlin. This standard dilution water is used for both, the maintenance of the test animals and the preparation of stock and test solutions of the test substance.

Pretreatment oe the test substance
To give the desired series oftest concentrations, the test substance was added to 1 litre of dilution water for each test concentration, then treated with an ultra turrax for 60 seconds at 8000 rpm and afterwards stirred for 24 hours on a magnetic stirrer. Finally undissolved particles of the test substance were removed by filtration using folded filters of pore size 7-12 Ilm.

Maintenance and Acclimatisation: A population of parthenogenetic females of synchronized age structure is maintained since more than 15 years in the test facility under constant temperature conditions (20 +/- 1°C) at a 16 : 8 hour light-dark photoperiod (illumination: < 1000 lux). The culture water (so-called 'M4 medium') is partly renewed once a week. The Daphnia are exclusively fed with unicellular green algae (Desmodesmus subspicatus) 'ad libitum'. Mortalities of parent Daphnia during the culture period are recorded daily in a semiquantitative way. The neonates are separated from their parent Daphnia by filtration prior to the acute test.

The results are expressed in terms of effective loadings (EL) at 24h and 48h. Considering the heterogeneous composition ofthe test substance, Water Accommodated Fractions (= WAFs) were tested. The results of the TOC determinations reflect a rough impression only of exposure conditions over time. No informations are available on a potential correlation between effective loadings and measured TOC values.

Observations were made on the swimming ability and the immobilisation rate, respectively, after 24 and 48 hours of exposure. Under the study conditions, the following values were determined: Highest test concentration resulting in 0% immobilisation (EC 0 48 h): 25 mg/L. Lowest test concentration resulting in 100% immobilisation (EC 100 48 h): > 100 mg/L. 40% and 90% immobilization was observed after 48 h at loading rates of 50 and 100 mg/L respectively. The results are expressed in terms of effective loadings (EL) at 24 h and 48 h. Under the study conditions, the nominal EL50 of the test substance in Daphnia magna was determined to be 58 mg/L (Bruns, 2002).
