Details on preparation and application of test substrate:
- Method of mixing into soil (if used): Test substance for the total amount of substrate was made up with 10 mL acetone for each exposure concentration. A weighed sub-sample of approx. 100 g wet weight was then taken from each treatment substrate and placed in a weighed container. The test solutions were spread out in vessels and left for approx. 1 hour to allow the solvent to evaporate. The sub-sample was reweighed to determine moisture loss over the evaporation period and any weight loss was replaced with reverse-osmosis (RO) water, where necessary. The sub-sample of test substrate was added to the respective batch of test substrate and thoroughly mixed by hand. RO water was then added to increase the moisture content to the required level (approx. 35% of dry weight).

EFFECT PARAMETERS MEASURED (with observation intervals if applicable): Survival of earthworms was assessed at days 7 and 14, missing earthworms were considered dead, due to rapid decomposition under test conditions. Any observed behavioural (i.e. burrowing time at day 0) or pathological symptoms and morphological changes were also recorded. After assessment at day 7 the test substrate and earthworms were replaced. The vessels were then re-weighed. Prior ot the day 14 earthworm assessment, each vessel was re-weighed to assess comparative moisture loss.

The percentage weight change at day 14 was analysed using one-way analysis of variance (ANOVA). Levene's test for equality of variances among
the dose levels was performed. Where this showed no evidence of heterogenicity (P ¿ 0.01), pair-wise comparisons with control were made using two-sample t-tests.
Treatment with test substance did not result in any statistically significant changes (P ¿ 0.01) in mean weight compared to control.

Conclusions:
The 14 day LC50 of Tri (hexyl, octyl, decyl) citrate to Eisenia foetida was observed to be > 1000 mg/kg, the highest dose tested.
The no observed effect concentration (NOEC) after 14 days of exposure was calculated to be 1000 mg/kg, based on earthworm weight.
There were no sub-lethal morphological changes observed in the test earthworms. There was not considered to be any evidence of treatment-related repellent effects.
The chloracetamide toxic reference results indicated that the earthworms from the source batch used for the test were responding normally in the test system.
Executive summary:

The acute toxicity of the read-across substance Tri (hexyl, octyl, decyl) citrate to the earthworm Eisenia foetida was determined during a 14 day exposure period according to OECD Guideline of Testing of Chemicals No. 207, adopted April 1984. For the combined range finding and limit test, the test substance was mixed with the OECD recommended artificial soil at nominal concentrations of 0.1, 1.0, 10, 100 and 1000 mg/kg dry substrate. After 7 and 14 days the effects of the test substance on the earthworms` survival (and any observable sub-lethal effects) were examined. The results were used to calculate LC50 values and no observed effect concentations.

The 14 day LC50 of the test substance to Eisenia foetida was observed to be > 1000 mg/kg, the highest dose tested. The NOEC for the test substance after 14 days of exposure was calculated to be 1000 mg/kg, based on a comparison of surviving earhworm weight changes. As there was no observable toxic effects at the 1000 mg/kg treatment rate, no further testing was carried out.

A definitve toxicity test with analytical grade chloroacetamide was carried out under similar conditions used for the definitive test with Tri (hexyl, octyl, decyl) citrate, using the eathworms from the same source and batch number. The 14 day LC50 of chloroacetamide to Eisenia foetida was calculated to be 32.84 mg/kg. Based on published data this is considered to be an acceptable response and indicated that earthworms from the source used in the test were responding normally in the test system.
