Description of key information

One key study and two supporting studies were identified to determine the potential for irritation due to exposure of 2-nitropropane when topicclay applied to rabbit skin.

Additional information

In the key study, the test was conducted in six albino rabbits on intact and abraded skin. A 0.5 ml sample of the skin material was applied to the shave backs of the rabbits and wrapped with an occlusive bandage for 24 hours. All erythema and edema scores at 24 and 72 hours were 0. Therefore the material was non-irritating. A similar study was done to determine the corrosion potential of 2 -nitropropane. Following a four-hour exposure period, the patches were removed and evaluated for erythema, edema and tissue destruction at 4, 24, and 48 hours after exposure. Results indicated that 2 -nitropropane is not corrosive. The last study was conducted on rabbits where the duration of treatment was once per day for five days. The treatment was applied unoccluded without any external sings of toxicity or irritation. The key study regarding the eye irritation potential of 2 -nitropropane was conducted on 6 rabbitsl administering 0.1 of test material into one eye with the other eye serving as control. The eyes were examined 24, 48, and 72 hours after treatment and scored by the Draize method. The mean eye irritation score was 1.0 +/- 1.7 at 24 hours and 0.3 +/- 0.8 at 48 hours and 0 at 72 hours.

Justification for classification or non-classification

Skin irritation studies show that 2 -nitropropane is non-irritating to intact and abraded rabbit skin and is therefore not classifiable under GHS.

Eye irritation studies, although the some response was seen in the animals there is not sufficient evidence to classify this substance as an eye irritant under GHS.
