Since the test substance is poorly water soluble, a saturated solution was prepared by shaking a nominal load of 1000 mg/L with deionized water for 24 hours, followed by membrane filtration.

Newborn daphnia (aged 0 to 23 hours, bred in the LAUS GmbH according to EN ISO 6341, originally obtained from Federal Environment Agency Berlin) were transferred to M4 Elendt medium for a settling-in period of 30 minutes. Undamaged individuals were caught using a glass tube (diameter 8 mm) and put into the respective test vessel (glass beakers, 100 mL) containing the test solution (loading 80 mL/animal, animals were held individually, 10 replicates/treatment, no aeration).
Following nominal concentrations were tested: 0, 10, 32, 100, 320 and 1000 mg/L (dilution water: tap water, TOC 0.82 mg/L, pH 8.17, conductivity 249 µS/cm, hardness 0.943 mmol/L; enriched to M4 Elendt medium).
Test vessels were inspected for offspring daily. Newborn daphnids were counted and removed from the test vessel.
Medium renewal was 3 x per week using freshly prepared test solutions.
pH and oxygen were measured in both the old and the new test solutions, hardness was determined once a week.
Daphnids were fed with Desmodesmus subspicatus (0.15-0.20 mg org. C/animal/day).
Statistical significance was tested by means of t-test or Weir-test, if homogeneity of variances was not given. EC50 was calculated according to a sigmoidal fit of the data (OriginTM).

Temperature during the test was 19.7 to 22.4°C, pH was 7.7 to 8.5, hardness was 157-283 mg CaCO3/L and O2 content was 7.7 to 9.4 mg/L. Lighting was below 2000 lux.
The NOEC was 32 mg/L, the corresponding LOEC 100 mg/L and the EC50 680 mg/L, based on nominal concentrations.
Mortality in the control was 0%, mean offspring was 96.
Following mean offspring numbers (in brackets inhibition of reproduction compared to the control) were obtained for the tested concentrations: 10 mg/L 104 (-9%), 32 mg/L 100 (-5%), 100 mg/L 74 (22%), 320 mg/L 82 (14%) and 1000 mg/L 30 (69%).
Mortality was 10% in the 320 mg/L treatment and 40% in the 1000 mg/L treatment. Results for the treatments 100 and 1000 mg/L were statistically significant (p<0.01).
