- Executive summary:
The 48–hour-acute toxicity of 2-propenoic acid to Daphnia magna was studied under static conditions. The study was conducted in accordance with EU method C.2 and OECD guideline 202, in compliance with GLP.
Daphnids were exposed to a control and nominal test concentrations of 10, 18, 32, 56 and 100 mg/L for 48 hours. Analytical measurements at 0 and 48 hour confirmed that the test concentration was not maintained within ± 20% of nominal. Immobilization and sub-lethal effects were observed daily. The test results are expressed in terms of geometric mean measured concentrations. The 48–hour EC50value was 47 mg/L. The 48–hour NOEC based on immobilization was 21 mg/L. There were no sub-lethal effects observed.
This study is classified as acceptable and satisfies the guideline requirements for an acute toxicity study with freshwater invertebrates.

Description of key information
The Key Study reported a 48 hour EC50 value for the toxicity of 2 -propenoic acid, 2 -fluoro, methyl ester to the freshwater invertebrate Daphnia magna of 47 mg/L based on geometric measured concentrations (Sacker, 2015).

The 24 and 48 hour median effective concentrations (EC50) of 2-propenoic acid, 2-fluoro, methyl ester to the freshwater crustaceanDaphnia magna(<24 hours old) were determined under static test conditions at a temperature of 20±1°C. The EC50 was defined as the concentration calculated from the data that resulted in immobilisation of 50% of theDaphniain the time period specified. Based on geometric mean measured test solution concentrations the 48-hour EC50 was 47 mg/L (no 95% CIs) and the 48-hour NOEC was 21 mg/L. No symptoms of toxicity (sub-lethal effects) were observed.
