A 28-day repeat dose rat inhalation study (6h exposure / 5 days per week) has been identified. Aside from the ocular effects in dogs, the liver and haematological system are the key target organs. In the identified inhalation study analyses were restricted to the respiratory tract, liver and haematological system. The leading toxicity was that of the liver, with the study identifying a dose response relationship for the liver and a NOAEC of 15 mg/m3. A route-to-route extrapolation from the dog oral NOAEL was not appropriate according to the ECHA guidance since famoxadone undergoes significant first pass metabolism (identified in the rat biliary study). Therefore it was determined that the inhalation DNEL should be derived from route specific data.
Assuming that inhalation absorption of famoxadone in the rat is equal to inhalation absorption in humans the following calculation is conducted:
Corrected NOAEC = 15 mg/m3 * (6h/d / 8h/d) * (6.7m3/10m3)
= 7.54 mg/m3

A 28-day rat dermal toxicity study was considered the most relevant study for dermal exposure assessment as it is route specific. In this study, famoxadone was applied to the skin of rats for 6hr/day for 28 consecutive days. The NOAEL was identified as 250 mg/kg bw/day for liver effects in males, and haematological effects in females.
Assuming that worst case (100%) dermal absorption of famoxadone in the rat is equal to dermal absorption in humans, and adjusting for daily and weekly durations of exposure, the following calculation is conducted:
Corrected NOAEL = 250 mg/kg bw/d * (6h/d / 8h/d) * (28d / 20d)
= 262.5 mg/kg bw/d
