Based on the geometric mean measured concentrations of the test item PU-2018-821, the 48 hours-
EC50 for Daphnia magna was 4.52 mg/L (95% confidence limits: 3.53 – 5.28 mg/L). - Executive summary:
The Daphnia magna acute immobilisation test was performed as a limit test.

In the acute immobilization test with Daphnia magna (STRAUS), the effects of the test item
PU-2018-821 (batch number: 2298695) were determined at the test facility according to OECD 202
(2004) from 2019-06-25 to 2019-06-27.
The study was conducted under semi-static conditions over a period of 48 hours with six
concentrations of the test item (nominal: 0.501 to 25.8 mg/L). They were prepared from a stock
solution of 25.8 mg test item/L in a geometric series with a separation factor of 2.2 between the test
concentrations. For details of the preparation, see section 4.2.
Twenty daphnids (divided into 4 replicates with 5 daphnids each) were exposed to each
concentration level and the control.
The concentrations of the test item PU-2018-821 were analytically verified via LC-MS/MS in the fresh
media at the start of the exposure and at the renewal of the test solutions (0 and 24 hours) as well
as in the 24-hours old media at the renewal and at the end of the test (24 and 48 hours) in all
concentration levels and the control. Details of the analytical method are presented in section 14.
The measured concentrations of the test item PU-2018-821 in the fresh media were in the range of
82 to 111% of the nominal values at the start of the exposure (0 hours) and at the renewal (24 hours).
The measured concentrations of the test item in the old media at the renewal and at the end of the
test (24 and 48 hours) were in the range of 54 to 98% of the nominal values. The analytical results
are presented in Table 5. The geometric mean measured concentrations of the test item
PU-2018-821 were 0.363 – 0.918 – 2.27 – 5.27 – 11.2 – 25.5 mg/L.
Since the measured concentrations did not stay within ± 20% of the initially measured concentrations
during the exposure, the ECx-values are based on the geometric mean measured concentrations of
the test item PU-2018-821. For details, see Table 1.
The validity criteria of the test guideline were fulfilled.
