- To produce the highest test concentration (1.2 mg/L) the test substance was added daily to 1 litre of dilution water, then treated with an ultra turrax for 60 seconds and afterwards stirred for 24 hours on a magnetic stirrer. Finally undissolved particles of the test substance were removed by filtration. The test concentrations 0.125, 0.25, and 0.5 mg/L were prepared daily by diluting a stock solution (0.5 mg/L) and treated in the same way as the test concentration 1 mg/L.

Maintenance and Acclimatisation: A population of parthenogenetic females of synchronized age structure is maintained since more than 15 years in the test facility under constant temperature conditions (20 +/- 1 °C) at a 16 : 8 hour light-dark photoperiod (illumination: < 1000 lux). The culture water (so-called 'M4 medium') is partly renewed once a week. The Daphnia are exclusively fed with unicellular green algae (Desmodesmus subspicatus CHODAT) 'ad libitum'. Mortalities of parent Daphnia during the culture period are recorded daily in a semiquantitative way. The neonates are separated from their parent Daphnia by filtration prior to the acute test. - Test type:

No. L383A, 29.12.92) Part C, Method 2 'Acute toxicity for Daphnia' which is in most parts equivalent to the OECD Guideline for Testing of Chemicals No. 202 'Daphnia sp., Acute Immobilisation Test and Reproduction Test, Part I - The 24h EC50 Acute Immobilisation Test'. The Daphnia were exposed to a range of concentrations, nominally 0.125, 0.25, 0.5 and 1 mg/L of test item dissolved in water. After 48 hours of exposure both an EC0 of 0.155 mg/L and an LC100 of 0.62 mg/L was measured. The geometric mean of the two values corresponding to the EC50 value is 0.31 mg/L. The results are expressed in terms of mean measured concentrations (at 48h). Measured concentrations ranged from 69.6 - 86% of nominal values at 0 hours, and from 45.6- 52% of nominal values at 48 hours, respectively. The hardness of the dilution water used was 15.0 °dH. This toxicity study is classified as acceptable and satisfies the guideline requirements for the acute daphnia study.
