- Three New Zealand White rabbits were supplied by David Percival Ltd, Moston, Sandbach, Cheshire, UK. At the start of the study the animals were in the weight range of 2.0 to 3.5 kg and were twelve to twenty weeks old. After an acclimatisation period of at least five days each animal was given a number unique within the study which was written with a black indelible marker-pen on the inner surface of the ear and on the cage label.
The animals were individually housed in suspended metal cages. Free access to mains drinking water and food (Certified Rabbit Diet (Code 5322) supplied by BCM IPS Limited, London, UK) was allowed throughout the study. The diet and drinking water were considered not to contain any contaminant of a level that might have affected the purpose or integrity of the study.
The temperature and relative humidity were set to achieve limits of 17 to 23°C and 30 to 70% respectively. Any occasional deviations from these targets were considered not to have affected the purpose or integrity of the study. The rate of air exchange was at least fifteen changes per hour and the lighting was controlled by a time switch to give twelve hours continuous light (06:00 to 18:00) and twelve hours darkness.
The animals were provided with environmental enrichment items which were considered not to contain any contaminant of a level that might have affected the purpose or integrity of the study.

- A volume of 0.1 ml of the test material, which was found to weigh approximately 72 mg (as measured by gently compacting the required volume into an adapted syringe.

- The upper and lower eyelids were held togetlter for about one second immediately after treatment, to prevent loss of the test material, and then released. The left eye remained untreated and was used for control purposes.
Assessment of ocular damage/irritation was made approximately 1 hour and 24, 48 and 72 hours following treatment.
Additional observations were made on Days 5, 7 and 14 to assess the reversibility of the ocular

application to the rabbit eye. The method was designed to meet the requirements of the following:

The albino rabbit has been shown to be a suitable model for this type of study and is
recommended in the test method. The results of the study are believed to be of value in predicting
the likely eye irritancy potential of the test material to man.

White residual test material was noted in all treated eyes during the study.
Diffuse corneal opacity was noted in one treated eye at the 24-hour observation with translucent
corneal opacity at the 48, 72-hour and 5-day observations. Diffuse corneal opacity was noted in
one other treated eye at the 72-hour observation with translucent corneal opacity at the 7-day
observation and opalescent corneal opacity at the 14-day observation. Vascularisation,
generalised ingrowth of vessels for 3 to 4 mm in length, was also noted in this treated eye at the
14-day observation.
Iridial inflammation was noted in one treated eye at the 24, 48, 72-hour and 5-day observations.
Iridial inflammation was noted in one other treated eye at the 72-hour, 7 and 14-day observations.
Moderate conjunctival irritation was noted in all treated eyes one hour after treatment. Severe
conjunctival irritation was noted in one treated eye, moderate conjunctival irritation was noted in
one other treated eye and minimal conjunctival irritation was noted in the remaining treated eye at
the 24 and 48-hour observations. Moderate conjunctival irritation was noted in two treated eyes
with minimal conjunctival irritation in the remaining treated eye at the 72-hour observation.
Moderate conjunctival irritation was noted in one treated eye at the 5-day observation. Severe
conjunctival irritation was noted in one treated eye with minimal conjunctival irritation in the
remaining treated eye at the 7-day observation. Severe conjunctival irritation persisted in one
treated eye at the 14-day observation.
Areas of haemorrhage along the edge of the nictitating membrane and the lower conjunctival
membranes was noted in one treated eye at the 24, 48, 72-hour and 5-day observations and in one
other treated eye at the 7 and 14-day observations. Pale appearance of the nictitating membrane
was noted in one treated eye at the 48, 72-hour and 5-day observations and in one other treated
eye at the 7 and 14-day observations. Blood stained discharge was noted in one treated eye at the
5-day observation.
One treated eye appeared normal at the 14-day observation.
Due to blood stained discharge and signs of pain and discomfort one animal was killed for
humane reasons in accordance with current UK Home Office guidelines on Day 5. Due to signs

- Executive summary:
The study was performed to assess the irritancy potential of the test material to the eye of the New Zealand White rabbit. The method was designed to meet the requirements of the following:
• OECD Guidelines for the Testing of Chemicals No. 405 "Acute Eye Irritation/Corrosion"
(adopted 24 April 2002)
• Commission Directive 92/69/EEC Method BS Acute Toxicity (Eye Irritation)
A single application of the test material to the non-irrigated eye of three rabbits produced translucent to opalescent corneal opacity, iridial inflammation and moderate to severe conjunctival irritation. Other ocular effects noted were vascularisation, with a generalised ingrowth of vessels for 3 to 4 mm in length, areas of haemorrhage on the nictitating and lower conjunctival membranes, pale appearance of the nictitating membrane and blood stained discharge. One treated eye appeared normal at the 14-day observation. Two animals were killed for humane reasons on Day 5 or 14, due to the severity of the reactions, in accordance with current UK Home Office guidelines.The test material produced a maximum group mean score of 40.5 and was classified as at least a severe irritant (Class 6 on a 1 to 8 scale) to the rabbit eye according to a modified Kay and Calandra classification system.
