- Remarks:
- Study has been performed according to OECD and/or EU guidelines and according to GLP principles.
- Qualifier:
- according to guideline
- Guideline:
- OECD Guideline 121 (Estimation of the Adsorption Coefficient (Koc) on Soil and on Sewage Sludge using High Performance Liquid Chromatography (HPLC))
- Deviations:
- no
- Qualifier:
- according to guideline
- Guideline:
- EU Method C.19 (Estimation of the Adsorption Coefficient (KOC) on Soil and Sewage Sludge Using High Performance Liquid Chromatography (HPLC))
- Deviations:
- no
- GLP compliance:
- yes (incl. QA statement)
- Type of method:
- HPLC estimation method
- Media:
- sewage sludge
- Radiolabelling:
- no
- Test temperature:
- 35°C
- Details on study design: HPLC method:
- According to the guidelines, the estimation of the Koc for test substances that are ionized for at least 10% within pH 5.5 to 7.5 should be performed with both the ionized and non-ionized form. Therefore, the pKa values of the test substance were calculated using the Perrin calculation method (pKalc 5.0, module in Pallas 3.0, CompuDrug International San Francisco, CA, USA). Based on the calculations (pKa -2.93 and -3.54), it was decided to perform the HPLC analysis at neutral pH.
Analytical conditions:
Instrument: Alliance Separation Module 2695 (Waters, Milford, MA, USA)
Detector: Refractive index detector 2414 (Waters)
Column: 150 mm x 4.6 mm i.d. Hypersil BDS-CN; dp = 5 µm (Thermo, San Jose, CA, USA)
Column temperature: 35 +/- 0.5°C
Mobile phase: 40/60 (v/v) methanol/Milli-Q water
Flow: 1 ml/min
Injection volume: 10 µl
RI detection: Sensitivity 64; time constant 1 second; temperature 35°C
A 3730 mg/l stock solution of the test substance was prepared in mobile phase.
The test substance blank solution was mobile phase.
The reference substance- and test substance solutions were injected in duplicate. Blank solutions were analysed by single injection. - Key result
- Type:
- Koc
- Value:
- 62 L/kg
- Temp.:
- 20 °C
- Key result
- Type:
- log Koc
- Value:
- 1.79 dimensionless
- Temp.:
- 20 °C
- Details on results (HPLC method):
- In the chromatogram of the test substance solution, one test substance peak was observed. The peaks at 2.2 and 5.4 minutes was also observed in the chromatogram of the blank solution. Therefore, it was assumed that these compounds did not derive from the test substance.
- Validity criteria fulfilled:
- yes
- Conclusions:
- The Koc and log Koc values of Ditetrahydrofurylpropane at neutral pH are: 62 and 1.79, respectively.
