PRETREATMENT OF TEST ITEM:
A stock solution was prepared to give the desired series of test concentrations. To achieve this 1000.5 mg of the test item were added to 10 litres of dilution water, treated for 1 h in an ultrasonic bath and afterwards stirred for 24 h on a magnetic stirrer.
Test organisms (species):
Danio rerio (previous name: Brachydanio rerio)
Details on test organisms:
- supplier: Aqua KlöGer (Germany)
- mean standard length (n=14): 3.16 cm (S.D. = 0.23 cm)
acclimatisation: - temperature: 20 - 24 °C; - dissolved oxygen: > 5 mg/l; - medication: none; - mortalities during acclimatization: < 5 % ; - feeding: Commercial fish food, daily. Feeding discontinued 24 h prior to test start; - Stock held since 2004-07-17 and acclimatised to the test conditions since then

Test temperature:
22.7 - 23.5 °C (daily monitoring)
pH:
7.6 - 7.9 (daily monitoring)
Dissolved oxygen:
7.8 - 8.6 (daily monitoring)
Nominal and measured concentrations:
nominal concentration: 25, 50 and 100 mg/L
Details on test conditions:
TEST SYSTEM:
- keeping water: synthetic fresh water according to ISO 7346
- glass aquaria holding 5 L of test media
- experimental design: 3 test concentrations plus 1 control; 10 animals per test concentration; no feeding during the exposure period; static system
- photoperiod: 16 hours light, 8 hours dark
- temperature: 20 - 24 °C +/- 1 °C
- aeration: gentle aeration via narrow glass tubes
- method of administration: stock solution
- solubilizer or other auxiliaries: ultrasonic bath, magnetic stirrer
- test concentration: 25, 50 and 100 mg/L plus one control
- Method of initiation: animals were placed in prepared media
- test parameter: mortality (recorded daily)
- Loading: 0.65 g body weight (wet weight) per litre
- medium renewal: none
Reference substance (positive control):

Effect conc.:
50 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mortality (fish)
Details on results:
All results are expressed in terms of nominal concentrations. Recovery rates ranges from 103.6 - 105.2% of nominal values at 0 hour, from 100.7 - 104.1% of nominal values at 24 hours, from 98.5 - 102.6% of nominal values at 48 hours and from 96.9 - 102.3% of nominal values at 72 hours and at 96 hours, respectively.
Validity criteria fulfilled:
yes
Remarks:
(-The mortality in the controls <10 % by the end of test. -The dissolved oxygen conc > 60%of the air-saturation value during study. -The pH did not vary by more than 1 unit.- substance concenteration > 80% during test )
Conclusions:
After 96 hours a LC 50 of 90.2 mg/L and a LC 100 of > 100 mg/L against fish was observed.
Executive summary:

In order to test acute toxicity to fish of the substance, Danio rerio was exposed to the test solution of 3 nominal concentration of the test substance ( 25, 50 and 100 mg/L) and blank control solution for a period of 96 h under static conditions.

Mortality and visible abnormalities were recorded at 2, 24, 48, 72 and 96 h. A LC 100 of > 100 mg/L and LC50 of 90.2 mg/L was observed after an exposure of 96h. This toxicity study is classified as acceptable and satisfies the guideline requirements for the toxicity study to fish (Danio rerio).
