The limit of quantitation (LOQ) for determination of DVS005u in the n-octanol phase by the method was defined as 800 mg/l, calculated as the product of the concentration of the lowest-level calibration standard (0.0400 mg a.i./l) and the method dilution factor for the n-octanol reagent blank (20000).

The LOQ for determination of DVS005u in the aqueous phase was defined as 0.500 µg/l, calculated as the product of the concentration of the lowest-level claibration standard (0.0100 mg/l = 10 µg/l) and the method dilution factor for the aqueous phase matrix blank sample (0.0500).

The mean calculated n-octanol/water partition coefficient for DVS005u at 25°C was 4.42E + 06, +/- 2.42E + 06 (CV = 55%). The corresponding log10 Pow was calculated at 6.58 +/- 0.32 (CV = 4.8%).
Executive summary:

The n-octanol / water partition coefficient (Pow) of DVS005u was determined in HPLC-grade reagent water at 25°C using the generatir column method. Analytical measurements of DVS005u concentrations in elution water were obtained by HPLC with UV detection. The DVS-005u concentration in the n-octanol phase used to coat the generator column was determined by an initial volumetric dilution in acetonitrile (ACN) and followed by a secondary volumetric dilution in 3:1 acetonitrile/water (v/v) dilution solvent into the calibration range of the HPLC/UV methodology. DVS005u concentration =s in the aqueous (column effluent) phase were determined by salting the aqueous phase with sodium chloride and extraction into dichloromethane (DCM), concentration if the extracts to near dryness, reconstitution with ACN and dilution with reagent water for a final composition of 3:1 ACN:water (v/v).
