Reference
Endpoint:
short-term toxicity to fish
Type of information:
experimental study
Adequacy of study:
key study
Study period:
26 - 30 May 1998
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
guideline study with acceptable restrictions
Remarks:
Undissolved test material is not removed from the test vessel. However, this is considered to be negligible as no mortality is observed in the treatment and the control throughout the test period. No analytical purity of the test substance is given.
Qualifier:
according to guideline
Guideline:
OECD Guideline 203 (Fish, Acute Toxicity Test)
Version / remarks:
adopted in 1992
Deviations:
no
Qualifier:
according to guideline
Guideline:
EU Method C.1 (Acute Toxicity for Fish)
GLP compliance:
yes
Analytical monitoring:
no
Vehicle:
no
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: The test solution of 100 mg/L was prepared by the addition of an appropriate amount of test substance directly to 20 L of dilution water in the test vessel.
- Evidence of undissolved material: The test solution was observed to be clear and colourless with an oil film on the surface.
Test organisms (species):
Oncorhynchus mykiss (previous name: Salmo gairdneri)
Details on test organisms:
TEST ORGANISM
- Common name: rainbow trout
- Source: obtained from West Country Trout, Trafalgar Farm, Boscastle, Cornwall, UK on 5 May 1998
- Length at study termination (mean, control): 4.7 (4.3 - 5.1 cm)
- Weight at study termination (mean, control): 1.4 (1.0 - 1.9 g)
- Method of breeding: Fishes were held in stock tanks. The fish showed no evidence of disease during holding, therefore no medication was necessary.
- Feeding during test: no

ACCLIMATION
- Acclimation period: Fishes were acclimatized to the test temperature (15 ± 1 °C) for a minimum period of 7 days.
- Acclimation conditions: same as test
- Type and amount of food: appropriate amounts of commercial fish food
- Feeding frequency: The frequency of feeding was not stated. Food was withheld from the fish for at least 24 h prior to the commencement of the test.
- Health during acclimation: No mortalities, other than 4 fish that had jumped from the holding tank, were observed in the fish in the 7 days prior to the start of the study.
Test type:
static
Water media type:
freshwater
Limit test:
yes
Total exposure duration:
96 h
Hardness:
44.7 mg/L as CaCO3
Test temperature:
15 ± 1 °C
pH:
7.4 - 7.6
Dissolved oxygen:
9.7 - 9.8 mg/L
Nominal and measured concentrations:
Nominal: 100 mg/L
Details on test conditions:
TEST SYSTEM
- Test vessel: not specified
- Material: glass; Fill volume: 20 L
- Aeration: The test vessels were gently aerated during the test period.
- No. of organisms per vessel: 10
- No. of vessels per concentration: 1
- No. of vessels per control: 1

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water: The dilution water was dechlorinated tap water supplied from a 100 m3 reservoir with a mean retention time of 24 h. It was passed through activated carbon, coarsely filtered to remove particulate material and dechlorinated with sodium thiosulphate. The treated water was then held in a secondary reservoir with a capacity of 36 m3 and a mean retention time of 8 h. The water was then passed through an ultraviolet steriliser to a second set of filters of 25 and 10 µm respectively and then to a third storage tank of 13.5 m3. It was delivered via a ring circuit to a temperature controlled header tank in the test lab set to a nominal 15 °C.
- Chlorine: < 2 µg/L
- Alkalinity: 26.2 mg/L as CaCO3
- Conductivity: 221 µS/cm
- Intervals of water quality measurement: Daily measurements were undertaken throughout the test period for pH, dissolved oxygen concentration and temperature.

OTHER TEST CONDITIONS
- Photoperiod: 16 h light : 8 h darkness with 20 min transition periods

EFFECT PARAMETERS MEASURED: Mortality and symptoms of toxicity were recorded after 24, 48, 72 and 96 h test duration.
Reference substance (positive control):
no
Duration:
96 h
Dose descriptor:
LC50
Effect conc.:
> 100 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mortality (fish)
Details on results:
No mortality was observed in the treatment and the control throughout the test period.

Description of key information

No toxic effects up to the limit of water solubility (< 0.05 mg/L in aqua dest) for Oncorhynchus mykiss (OECD 203)

Key value for chemical safety assessment

Additional information

One study investigating the short-term toxicity of Propane-1,2,3-triyl 2-ethylhexanoate (CAS No. 7360-38-5) to freshwater fish is available. The GLP study was conducted under static conditions according to OECD guideline 203 using Oncorhynchus mykiss as test organism. A limit test was performed with a 100 mg/L test solution prepared by directly weighing the test substance into the test vessels without further processing steps. No mortality was observed in the treatment and the control throughout the test period of 96 h. Hence, a 96 h-LC50 is determined to be > 100 mg/L based on the nominal test concentration.
