Sampling and analysis

Analytical monitoring:
yes

Test solutions

Vehicle:
no
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: The test concentrations were prepared by dilution of a stock dispersion. The concentration of the stock dispersion was 111.1 mg/L. The test substance was weighed in and treated by ultra sonification for 30 minutes. The stock dispersion was a dispersion with fine particles, which were homogenously distributed, so that the stock dispersion could be used to prepare the test batches.

Test organisms

Test organisms (species):
Desmodesmus subspicatus (previous name: Scenedesmus subspicatus)
Details on test organisms:
TEST ORGANISM
- Common name: unicellular green alga
- Strain: SAG 86.81
- Source (laboratory, culture collection): obtained at regular intervals from SAG (Collection of algal cultures in Göttingen) and kept in liquid culture in the Laboratory of Experimental Toxicology and Ecology
- Age of inoculum (at test initiation): 3 days

PRE CULTURE
- Seed-culture: A seed culture was incubated for 4 days at 23 °C ± 2 °C
- Final cell density: 150E+04 cells/ml

- Pre-culture: Seed-culture was taken to inoculate a pre-culture (initial cell density: 1 x 104 cells/mL). The pre-culture was
incubated for 3 days at 23 °C ± 2 °C.
- Final cell density: 92E+04 cells/ml

- No. of vessels per control (replicates): 3

GROWTH MEDIUM
- Standard medium used: yes

TEST MEDIUM / WATER PARAMETERS
- Intervals of water quality measurement: temperature was continuously measured during the whole test period in the climate
chamber; pH in additional uninoculated parallels: after 0 h in parallel 5 (this parallel was disposed of after measurement), after 72 h in parallel 6 and in the inoculated parallel 1 of each concentration.

OTHER TEST CONDITIONS
- Photoperiod: permanent illumination
- Light intensity and quality: Artificial light, type universal white, about 6000 – 10000 Lux at a wave length of 400 - 700 nm

EFFECT PARAMETERS MEASURED:
- Determination of cell concentrations: after 72 h in a counting chamber (Neubauer improved) in replicate No. 2 of the inoculated control
- Chlorophyll measurement: In vivo chlorophyll-a-fluorescence (pulsed excitation with light flashes having a wavelength of 435 nm) after 0, 24, 48 and 72 h

TEST CONCENTRATIONS
