The acute toxicity of 2 -Hexyl-2 '-hydroxy-5 'methyldecananilide (AF-654) to aquatic invertebrates was determined in a study by de Roode (2004). This test was conducted according to EU Method C.2 and OECD Guideline 202 following GLP requirements. During the study, two limit tests were performed under static conditions. Daphnids (Daphnia magna) were exposed for 48 hours to a filtered solution prepared at 100 mg/L nominal concentration. A verification of test concentration was performed by HPLC analysis. No immobility or other adverse effects could be observed at the end of the exposure period. Therefore, an EC50 value could not be calculated and it is considered to be > 0.017 mg/L (measured concentration). In addition, an EC50 > 100 mg/L based on the nominal test concentration of the test substance in a filtered solution is assumed and reported in the study summary.

In conclusion, AF-654 was not acutely toxic to aquatic invertebrates under the tested conditions. No effects of the substance were observed up to the water solubility limit of the test substance (< 0.02 mg/L).
