The acute toxicity to Daphnia magna was carried out according to OECD 202 Daphnia sp., Acute Immobilisation Test and EU Method C.2 guidelines under GLP. Following preliminary range finding/limit test in the 0.1 to 100 mg/L range, an identically conducted definitive study was completed in 100 mL closed glass flasks with a 80 mL fill volume under static conditions.Preparation of test solutions started with individually prepared loading rates that were magnetically stirred for two days. The resulting suspensions were left to settle for an hour, were after the Water Accommodated Fractions (WAFs) were siphoned from the middle of the settled suspensions. The WAFs were all clear and colourless and were used for testing. Preparation of test solutions started with the highest loading rate of 100 mg/L (combined limit/range-finding test) or 10 mg/L (final test). The definitive test WAF loading rates were 0 (control), 0.46, 1.0, 2.2, 4.6 and 10 mg/L. The total exposure period was 48 hours and samples for analytical confirmation of actual exposure concentrations were taken at the start and at the end of the test. Analysis of the samples taken at the start of the final test showed measured concentrations of 0.37, 1.0, 1.9, 4.8 and 10.0 mg/L in the WAFs prepared at 0.46, 1.0, 2.2, 4.6 and 10 mg/L, respectively. Concentrations measured after 48 hours decreased by approximately 20 to 30% below the initial measured concentrations. The range tested based on average exposure concentrations corresponded to 0.32, 0.88, 1.6, 4.2 and 9.0 mg/L. The study met the acceptability criteria prescribed by the protocol and was considered valid. The results demonstrated 45% immobility at 4.2 mg/L and 100% immobility at 9.0 mg/L average exposure concentration. 95% confidence intervals could not be determined. Under the conditions of this study, the 48h-EC50 based on geometric mean measured average exposure concentrations was 4.3 (C.I. – ) mg/L.
