Blacksburg, Virginia municipal tapwater was dechlorinated using activated carbon, deionized using mixed bed deionization tanks to a resistivity of > 1 megohm/cm, and polished with a Millipore, Milli Q UV Plus and 0.2 micron filter to a final resistivity of > 18 megohm/cm. This is referred to as BMI DI water and was used to prepare Hard Reconstituted Water following EPA guidelines (e.g., EPA/600/4-90/027F). Evian spring water was diluted with BMI DI water to make the HRW with a final hardness of 166 mg/L.

Daphnids (Daphnia magna) were used in this study. These organisms were obtained from in-house cultures. Gravid adults were isolated less than 24 hours before the test start date. Offspring to be used were then harvested on the morning of the test.

Range-Finding Tests
A 48h static range-finding acute test was performed to estimate the range of concentrations for use in the definitive testing. Range-finding tests were conducted using five (5) organisms per chamber of each concentration. D. magna <24 h old were exposed to a logarithmic series of product concentrations in HRW. The organisms were loaded with plastic 5 ml pipets.

The highest nominal concentration used was 1000 mg/L. Test water physicochemical parameters were recorded daily during the 48-hour study. Live and mobile daphnids were counted daily and recorded. The daphnids were considered dead and/or immobile if there was no visible movement and if there was no reaction to touching.

Definitive Tests
Definitive testing was conducted based on the results of the range-finding test. Four replicates of five compound treatments and a control were prepared. Twenty randomly selected daphnids were exposed to each treatment and the control for 48 hours. The tests were performed in 50 ml polyethylene beakers containing 35 ml of test solution each. D. magna individuals were not fed for the test duration and the test temperature was 20 + 1°C.

Conductivity was measured in the treatments and the control at the beginning and end of the study. Temperature, dissolved oxygen, pH, and number of live organisms were recorded daily. Alkalinity and hardness were determined for the dilution water at the beginning of the study.

Validity of the Test
In the control, no more than ten (10) percent of the organisms were immobilized (Table 3) and the dissolved oxygen concentration at the end of the test was more than 3 mg/L in both the control and test vessels (Appendix B). The test was valid.

Sample recovery ranged from 101.3% (20 mg/L) of the nominal concentration to 112% (1.25 mg/L) of the nominal concentration. Since the measured concentrations were within 20% of the nominal, nominal concentrations were used in determining the EC50.
