- Any observations (e.g. precipitation) that might cause a difference between measured and nominal values: During the exposure period the measured concentrations decreased in the range finder and definitive test from nominal values. No observations were made to account for the difference.
- Effect concentrations exceeding solubility of substance in test medium: No. - Results with reference substance (positive control):
- - Results with reference substance valid?: Yes.
- EC50: The EC50 for growth rate reduction (ERC50: 0-72h) was 1.2 mg/l with a 95% confidence interval ranging from 1.0 to 1.3 mg/l. The EC50 for yield inhibition (EYC50: 0-72h) was 0.52 mg/l with a 95% confidence interval ranging from 0.45 to 0.62 mg/l. The results from the positive control with potassium dichromate were within the normal ranges for this reference item.
- Other: Exposure conditions and data evaluation for the positive control were similar to those in the definitive test. - Reported statistics and error estimates:
- Statistical analysis of the growth rate and yield data was carried out for the control and all test concentrations using one way analysis of variance incorporating Bartlett's test for homogeneity of variance (Sokal and Rohlf, 1981) and Dunnett's multiple comparison procedure for comparing several treatments with a control (Dunnett, 1955).
Growth rate and Yield: There were no statistically significant differences between the control, 1.3 and 4.4 mg/L test concentrations (P≥0.05), however all other test concentrations were significantly different (P<0.05) and, therefore the "No Observed Effect Concentration" (NOEC) based on growth rate was 4.4 mg/L. Correspondingly the "Lowest Observed Effect Concentration" (LOEC) based on growth rate was 14 mg/L. - Validity criteria fulfilled:
- yes
- Conclusions:
- The test item 72h-EC50 (growth rate reduction) was 12 (95% CL: 10 – 14) mg/L based on geometric mean measured concentrations. The equivalent EC10 was 5.4 mg/L. The NOEC was 4.4 mg/L.
- Executive summary:
The algal growth inhibition to Pseudokirchneriella subcapitata, was carried out according to OECD 201 Freshwater Alga and Cyanobacteria, Growth Inhibition Test and EU Method C.3 guidelines. Following a preliminary range-finding test, P.subcapitata was exposed to solutions of the test item at nominal concentrations of 1.0, 3.2, 10, 32 and 100 % (v/v) saturated solution (three replicate flasks per concentration) for 72 hours, under constant illumination and shaking at a temperature of 24 ± 1 °C. The test item solutions were prepared by stirring an excess (500 mg/L) of test item in culture medium using a propeller stirrer at approximately 1500 rpm for 24 hours. After the stirring period any undissolved test item was removed by filtration (0.2 μm Sartorius Sartopore filter, first approximate 2 litres discarded in order to pre-condition the filter) to produce a 100 % (v/v) saturated solution of the test item. This saturated solution was then further diluted as necessary, to provide the remaining test groups. Samples of the algal populations were removed daily and cell concentrations determined for each control and treatment group. Analysis of the test preparations at 0 hours showed measured test concentrations to range from 1.9 to 170 mg/L. A decline in measured test concentration was observed at 72 hours to between 0.89 and 112 mg/L (47 % to 66 % of the 0-Hour measured test concentrations) and hence it was considered appropriate to calculate the results based on the geometric mean measured test concentration. The study met the acceptability criteria prescribed by the guideline. The test item 72h-EC50 (growth rate reduction) was 12 (95% CL: 10 – 14) mg/L based on geometric mean measured concentrations. The equivalent EC10 was 5.4 mg/L. The NOEC was 4.4 mg/L.

Additional information
Key study : OECD TG 201, 2013 : Read-Across - SOURCE (4-methyl-2-phenyltetrahydro-2H-pyran): The algal growth inhibition to Pseudokirchneriella subcapitata, was carried out according to OECD 201 Freshwater Alga and Cyanobacteria, Growth Inhibition Test and EU Method C.3 guidelines.
