All loading rates and the control were analytically verified via GC MS at the start (0 hours) and at the end of the exposure (72 hours) with algae.
At the start, samples were taken from one additional replicate of the test item loading rate and the control and at the end samples were taken from pooled replicates.

PREPARATION AND APPLICATION OF TEST SOLUTION
A saturated solution with a nominal loading rate of 10.0 mg test item/L was prepared once at room temperature 96 ± 1 hour prior to the start of the exposure. An appropriate amount of the test item was weighed out. The test item was applied onto a glass slide. The glass slide with the test item was inserted into a glass bottle with an appropriate amount of dilution water (see Table 2). The saturated solution was stirred for 96 ± 1 hour (1100 rpm, room temperature). After a separation phase of 1 hour (at room temperature), the saturated solution was removed by siphoning (from the approximate middle of the glass flask). The saturated solution was be checked via laser beam (Tyndall effect) for undissolved test item. The Tyndall effect was negative.

Dilution levels of the saturated solution were tested in a geometric series with a dilution factor of 1.78: 9.96 - 17.7 - 31.6 - 56.2 - 100% of the saturated solution, corresponding to the geometric mean measured test item concentration of 11.9 – 22.6 – 39.7 – 70.6 – 119 µg/L. The dilution levels are based on the results of a preliminary range finding test (non-GLP).

The control solution was prepared without test item following the same method as specified for the saturated solution. Six replicates (without test item) were tested under the same test conditions as the test item replicates. Potassium dichromate was tested as a reference item

The study was performed as a static test in open vessels under culture conditions. The flasks contained 100 mL either reconstituted water or test medium (test item group).
The duration of exposure was 72 hours. The algal biomass in each flask was determined daily during the exposure period via Chlorophyll a-fluorescence.
Microscopic evaluation of the cells at the start and at the end of the incubation period was carried out.

Results with reference substance (positive control):
The toxicity of potassium dichromate (SIGMA ALDRICH, batch number MKBV0900V, purity 99.0%, CAS RN 7778-50-9) to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined over a period of 72 hours from 2020-04-20 to 2020-04-23. The reference item toxicity is in the valid range which was established by calculation of the average of the historic reference data since 2006, and the limits were set using the threefold standard deviation of these values.
ErC50 = 1.12 mg/L, EyC50 = 0.59 mg/L
Reported statistics and error estimates:
The EC50-values for inhibition of growth rate (ErC50) and yield (EyC50) after 72 hours above 119 μg/L and thus above the water saturation limit concentration of the pure test material, respectively.

The EC50 and EC10 value (based on growth rate) of the test material is thus clearly above the water solubility of the pure substance corresponding to a nominal concentration of 100 mg test item/L (cf. OECD 201).
Executive summary:

The toxicity of the test material to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined according to the principles of OECD 201 and Council Regulation (EC) No. 266/2016/Method C.3.
The aim of the study was the determination of the effects on growth rate and yield over a period of 72 hours.

The study was conducted under static conditions with an initial cell density of 5375 cells/mL. A saturated solution with a nominal loading rate of 10.0 mg test item/L was prepared once 96 ± 1 hour prior to the start of the exposure. Description of key information

In the OECD 201 assay performed for this compound, no effects on Daphnia magna were observed up to the saturated solution of the test item.

The EC50 and EC10 value for inhibition of growth rate after 72 hours were > 119 mg/L (geometric mean measured) corresponding to a nominal concentration of 100 mg/L.
