In Article 13 of Regulation (EC) No 1907/2006, it is laid down that information on intrinsic properties of substances may be generated by means other than tests, provided that the conditions set out in Annex XI (of the same Regulation) are met. Therefore, and for reasons of animal welfare further experimental studies on aquatic toxicity are not provided.

 According to Annex XI of Regulation (EC) No 1907/2006 (Q)SAR results can be used if (1) the scientific validity of the (Q)SAR model has been established, (2) the substance falls within the applicability domain of the (Q)SAR model, (3) the results are adequate for the purpose of classification and labeling and/or risk assessment and (4) adequate and reliable documentation of the applied method is provided.

 For the assessment of the reaction mass of 4-methoxyphenol and 4-methoxy-cyclohexanone and 4-methoxy-cyclohexanol (EC 947-389-1; Mochon roh) (Q)SAR results were used to assess the acute toxicity to fish. The criteria listed in Annex XI of Regulation (EC) No 1907/2006 are considered to be adequately fulfilled and therefore the endpoint(s) sufficiently covered and suitable for risk assessment.

 

Assessment:

No experimental data on the short-term toxicity to fish of Mochon roh are available. In order to assess the short-term toxicity to fish a read across approach has been developed using data for its three constituents.

In a GLP OECD 203 study, conducted with 4-methoxycyclohexanone (CAS 13482-23-0), a 96 -h LC50 value of more than 100 mg/L based on nominal concentrations was determined (analytically verified, ECHA, disseminated data). Therefore, the substance is considered to be acutely not harmful to fish.

Mequinol (CAS 150-76-5) was tested according to OECD 201 in a flow through system. The 96 -h LC50 value was estimated to be 28.5 mg/L (Hodson, 1984, published: Environ Toxicol Chem, 3 (2)). The latter LC50 was determined based on measured test item concentrations. Therefore, Mequinol is acutely harmful to fish.

Moreover, LC50 values were calculated for 4-Methoxycyclohexan-1-ol (CAS 18068-06-9) with several estimation models. The substance is within the applicability domain of the applied models. Based on the model Fish Classification (SARPy/IRFMN) v1.0.2 (VEGA v1.1.3), the substance is classified as NON-Toxic (96 -h LC50 > 100 mg/L). ECOSAR v1.11 categorised the substance as a neutral organic. The 96 -h LC50 was estimated to be 1284 mg/L. The estimation model US EPA T.E.S.T v4.2.1 estimated the LC50 value to be 298 mg/L. Based on the available calculated data the substance is considered to be with high probability acutely not harmful to fish.

Reaction mass of 4-methoxyphenol and 4-methoxy-cyclohexanone and 4-methoxy-cyclohexanol (EC 947-389-1; Mochon roh) is a mixture of 4-methoxycyclohexanone (CAS 13482-23-0), 4-Methoxycyclohexan-1-ol (CAS 18068-06-9), Mequinol (CAS 150-76-5). Based on the available experimental and estimated data for its three constituents, Mochon roh is acutely harmful to fish.
