Description of key information

Short-term toxicity to fish

No study is available on The test item. Two studies from supporting substance (structural analogue or surrogate) are available.

 

In this static limit test according OECD 203 the test item Sodium-N-cocoyl glycinate (a.i content of solution 29.8% ww) did not cause any effects to zebrafish after 96h when tested with a nominal concentration of 100 mg/L (29.8 mg/L a.i.). LC0 and NOEC are 100 mg/L (29.8 mg/L a.i.). LC100 is > 100 mg/L (> 29.8 mg/L a.i.).

 

In a static test according OECD 203 the test item Sodium N-octoyl glutamate the LC50 of the test item was found to be 150 mg/L test item or 62.4 mg/L a.i.

Short-term toxicity to invertebrate

No study is available on the test item. Two studies from supporting substance (structural analogue or surrogate) are available.

 

Based on the nominal concentrations of the test item Sodium N-cocoyl glycinate the 48-h EC50 for Daphnia magna was estimated to be 33.0 mg/L (95 % confidence limits: 30.2 - 37.2 mg/L) or 6.5 mg/L based on active ingredient.

The NOEC after 48 h was determined to be 10.2 mg/L as the highest tested concentration level with no biologically significant effect. The LOEC after 48 h was determined to be 25.6 mg/L as the lowest tested concentration level with a biologically significant effect.

 

In the acute immobilization test with Daphnia magna the effects of the test item Sodium N-octoyl glutamate were determined according to the Guidelien Din 38 412 Part 1 and Part 11. Based on the nominal concentration of the test item the m48 h EC50 for Daphnia magna was estimated to be 18 mg/L based on test material.

Long Term Toxicity to invertebrate

The Lowest Observed Effect Concentration (LOEC) and the No Observed Effect Concentration (NOEC) for the reproductive output were determined directly from reduction of the reproductive output. EC10- and EC50- values for the reproductive output and the adult mortality could not be estimated because effects > 10 % were not observed.

As the concentrations in the old media (48 h) were below LOQ, the endpoints were assessed based on the nominal test item concentrations corrected with a factor of 2. Endpoint assessment based on mean measured concentrations is not possible, because the toxicity cannot be attributed to any single compound of the test item Hostapon SG containing different C-chains of the N-Cocoyl glycine sodium salt. Therefore, it was concluded that an evaluation on half of the nominal concentrations sufficiently describes a worst case scenario. Additionally the endpoints are given based on half of nominal concentrations of the active ingredient (a.i.) taking its content of 19.8 % into account.

Toxicity to aquatic algae

In this study Hostapon SG dried was found to inhibit the growth of the freshwater green alga Desmodesmus subspicatus after 72 h. The NOEC-values for inhibition of specific growth rate and inhibition of yield after 72 h were 32.0 and 1.00 mg/L, respectively. The LOEC-values for inhibition of specific growth rate and inhibition of yield after 72 h were 100 and 3.20 mg/L, respectively. The EC50-values for inhibition of specific growth rate (ErC50) and yield (EyC50) with 95 % Confidence Intervals after 72 h were 89.9 (84.3 – 96.2) and 30.5 (21.5 – 43.4) mg/L, respectively.

All effect levels are given based on nominal concentrations of Hostapon SG dried.Values based on a.i. basis are given in the result section.

 

No studies investigating the toxicity of Disodium N-lauroyl-glutamate to algae are available. Thus, available data for the source substance Sodium N-cocoyl-glycinate was used in a read across approach. The read across is considered valid as both substances are structurally identical (solely differing in the degree of deprotonation being di- and monosodium salts, respectively), which assume very similar physico-chemical properties resulting in identical environmental behavior and ecotoxicity.

The read across key study was performed according to GLP and OECD guideline 201 using Desmodesmus subspicatus as test organism. In this study, Hostapon SG dried was found to inhibit the growth of the freshwater green alga Desmodesmus subspicatus after 72 h. The NOEC-values for inhibition of specific growth rate and inhibition of yield after 72 h were 32.0 and 1.00 mg/L, respectively. The LOEC-values for inhibition of specific growth rate and inhibition of yield after 72 h were 100 and 3.20 mg/L, respectively. The EC50-values for inhibition of specific growth rate (ErC50) and yield (EyC50) with 95 % Confidence Intervals after 72 h were 89.9 (84.3 – 96.2) and 30.5 (21.5 – 43.4) mg/L, respectively.

Toxicity to microorganisms

No studies investigating the toxicity of Disodium N-lauroyl-glutamate to algae are available. Thus, available data for the source substance Sodium N-cocoyl-glycinate was used in a read across approach. The read across is considered valid as both substances are structurally identical (solely differing in the degree of deprotonation being di- and monosodium salts, respectively), which assume very similar physico-chemical properties resulting in identical environmental behavior and ecotoxicity.

A Respiration Inhibition Test with activated sludge according to OECD Guideline No. 209 was carried out with the test item Sodium N-cocoyl-glycinate. Test system was activated sludge of the municipal treatment plant of D-31137 Hildesheim. The test was carried out under static conditions with the nominal active ingredient concentrations 100 - 180 - 320 - 580 - 1000 mg/L. The respiration rates of the control, reference and test item replicates were measured after a contact time of three hours, and the inhibitory effects of the test and reference item were determined in comparison to the control respiration rates. The mean inhibition of respiration for the test item replicates ranged from -33 % to 62 %.

The NOEC of the active ingredient is 320 mg/L, the EC10 is 372 mg/L and the EC50 791 mg/L to activated sludge of a municipal sewage treatment plant.
