Short-term toxicity to fish:
Fish Acute Toxicity test according to OECD Guideline 203 was conducted forReaction mass of 3H-Indolium, 2-[2-[4-[(2-cyanoethyl)methylamino] phenyl]ethenyl]-1,3,3-trimethyl- & acetateon Zebra fish.
The nominal concentration selected for the experiment were6.25mg/L,12.5mg/L,25mg/L,50mg/L,100mg/LandZebra FishDanio reriowere exposed to these concentration for 96 hours.
The test substance was soluble in water. Therefore, the test solution was prepared by dissolving 1 g of the test substance in 1 liters of potable water (passed through reverse osmosis system) with continuous 1 hr stirring,After stirring stock was analytically detected ,the solubility found 848.46 mg/L.
Bowl aquaria containing 2 liters of potable water (passed through reverse osmosis system) were loaded with 8 fishes. A static procedure was used for the study and it was conducted in compliance with the OECD guideline 203.
The nominal concentration selected for the experiment were6.25mg/L,12.5mg/L,25mg/L,50mg/L,100mg/L ,respectivelyand Zebra fish were exposed to this concentration for 96 hours. The lethal concentrations LC50 was found to be 12.5 mg/l
After 96 hours of exposure to test itemReaction mass of 3H-Indolium, 2-[2-[4-[(2-cyanoethyl)methylamino] phenyl]ethenyl]-1,3,3-trimethyl- & acetateto various nominal test concentrations, LC50 was determine to be 12.5 mg/l . Based on the LC50, it can be consider that the chemical was toxic and can be consider to be classified as "Acute aquatic 2" as per the CLP classification criteria.

Data available for the structurally and functionally similar read across chemicals has been reviewed to determine the short term toxicity of aquatic invertebrate of the test chemicalReaction mass of 3H-Indolium, 2-[2-[4-[(2-cyanoethyl)methylamino]phenyl]ethenyl]-1,3,3-trimethyl- & acetate.The studies are as mentioned below:
1.Daphnia sp., Acute Immobilization Test according to OECD Guideline 202 was conducted for test material. The nominal concent ration selected for the experiment was 100 mg/L and test Daphnids were exposed to this concentration for 48hours. The Effective concentrations EC50 was found to be 100 mg/L. Also the substance was found to be not rapidly degradable.Thus according to CLP Criteria for aquatic classification of the substance, it is concluded that test material exhibits toxicity to aquatic invertebrate (Daphnia magna) in the chronic 3 classification category.
EC50 (48 hours) Experimental = 100 mg/L
2.Determination of the inhibition of the mobility of daphnids was carried out with the substance,according to OECD Guideline 202.
The test substance was tested at the concentrations 0, 6, 10, 17, 29, 50 and 100 mg/L. Effects on immobilisation were observed for 48 hours. The median effective concentration (EC50) for the test substance,1-(3-chloropropyl)-1,3-dihydro-2H-benzimidazol-2-one, in Daphnia magna was determined to be 29.8 mg/L for immobilisation effects.
This value indicates that the substance is likely to be hazardous to aquatic invertebrates and can be classified as Aquatic Chronic category 3 as per the CLP criteria.
Thus, based on the above summarised studies,Reaction mass of 3H-Indolium, 2-[2-[4-[(2-cyanoethyl)methylamino]phenyl]ethenyl]-1,3,3-trimethyl- & acetateand it’s functionally similar read across substance, it can be concluded that effect concetration value is in the range of 29 to 100 mg/

Toxicity to aquatic algae and cyanobacteria:
Data available for the structurally and functionally similar read across chemicals has been reviewed todetermine the toxicity of aquatic algae and cyanobacteria of the test chemical Reaction mass of 3H-Indolium, 2-[2-[4-[(2-cyanoethyl)methylamino]phenyl]ethenyl]-1,3,3-trimethyl- & acetate.The studies are as mentioned below.
1.The algal growth inhibition test was performed using as test material on Chlorella sp .
Chlorella sp were grown on 500mL Erlenmeyer flask.Algae were added into 100mL of test water containing 50, 100, 150, 200 and 250 mg/L of 73.6, 67.9, 33.3, 25.0 and 16.6% concentration of test chemical .
The flask was placed in an ambient temperature provided air and sealed with parafilm.Flasks were continuous illuminated (cool white fluorescent light approximately 120 mE/s/m2 or 6,000 lux at water level.
Determination of algal growth by measuring live cell count, Chlorophyll a, dry weight and absorbance at 720nm. The effect concentration of test chemical on Chlorella sp. was observed to be 10.8 mg/l.
2.Freshwater algal growth inhibition test was carried out on Desmodesmus subspicatus with the substance,according to OECD Guideline 201.
The test substance was dissolved in DMSO/ OECD growth medium and tested at the concentrations 0, 3, 6, 12, 24 and 50 mg/L. Effects on the growth rate of the organism were studied. The median effective concentration (EC50) for the test substance,diphenylacetonitrile,,in Desmodesmus subspicatus was determined to be 12.9 mg/L.
This value indicates that the substance is likely to be hazardous to aquatic algae and can be classified as Aquatic Chronic Category 3 as per the CLP criteria.
Thus, based on the above summarised studies,Reaction mass of 3H-Indolium, 2-[2-[4-[(2-cyanoethyl)methylamino]phenyl]ethenyl]-1,3,3-trimethyl- & acetateand it’s structurally and functionally similar read across substance, it can be concluded that effect concetration value is in the range of 10 to 12 mg/ L. A screening method based on the measurement of the respiration rate of activated sludge for assessing the possible inhibitory effect of dyestuffs on aerobic waste-water bacteria. The test principle involves measuring the respiration rate of an activated sludge and comparing it with the respiration rate of the same activated sludge under identical conditions, but in the presence of the chemical under test. The test was carried out in activated sludge respiration rate apparatus with constant 20 ± 2°C and pH about 7-8. The test concentration used was 100 mg/l. OECD recommended synthetic sewage was used as feed, while activated sludge was obtained from a sewage works treating predominantly domestic sewage or from a sewage works treating predominantly industrial waste water. The respiration rate of an activated sludge and the respiration rate of activated sludge with test chemical were noted down. In order to calculate the inhibitory effect of a particular chemical at 100 mg/l test concentration its respiration rate is expressed as a percentage of the mean of the two control respiration rates. Thus, IC50 value (concentration for 50% inhibition of respiration rate) for the test chemical on activated sludge (aerobic bacteria) is determined to be10 - 100 mg/1 after 3 hrs of exposure.
The test chemical Reaction mass of 3H-Indolium, 2-[2-[4-[(2-cyanoethyl)methylamino]phenyl]ethenyl]-1,3,3-trimethyl- & acetate is likely to be toxic to microorganism in the concentration range of 10-100 mg/l
