- Sampling method: 2 mL samples were taken after 0, 24 and 72 h. At the end of the exposure period, the replicates with algae were pooled at each concentration before sampling. Compliance with the quality criteria regarding maintenance of actual concentrations was checked by running a test vessel at the highest test item concentration but without algae and samples for analysis were taken at the start, after 24 h of exposure and at the end of the test period.

- PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: Preparation of test solutions started with a loading rate of 100 mg/L applying a three-day period of magnetic stirring to ensure maximum dissolution of the test item in medium. Thereafter, the aqueous Saturated Solution (SS) was collected by filtration through a 0.45 µm membrane filter (RC55, Whatman) and used as the highest test concentration. Lower test concentrations were prepared by subsequent dilutions of the SS in test medium. After preparation, volumes of 50 mL were added to each replicate of the respective test concentration.

- The EC50 for growth rate inhibition (72 h ErC50) was 1.1 mg/L. The historical ranges for growth rate inhibition lie between 0.82 and 2.3 mg/L. Hence, the 72 h ErC50 for the algal culture tested corresponds with this range.
- Reported statistics and error estimates:
- For determination of the NOEC and the EC50 the approaches recommended in the OECD guideline 201 were used. An effect was considered to be significant if statistical analysis of the data obtained for the test concentrations compared with those obtained in the negative control revealed significant inhibition of growth rate or inhibition of yield (STUDENT-t test for Homogeneous Variances, α=0.05, one-sided, smaller). No EC50-values could be calculated because the test item proved to be non-toxic (EC50 > maximum soluble concentration tested). The calculations were performed with ToxRat Professional v. 3.2.1. (ToxRat Solutions® GmbH, Germany).

- In conclusion, under the conditions of the present study with Pseudokirchneriella subcapitata, no significant inhibition of growth rate was recorded at any of the concentrations of 4,4’-[isopropylidenebis(4,1-phenyleneoxy)]dianiline tested.
The EC50 for growth rate inhibition (72 h ErC50) was beyond the range tested, i.e. exceeded a TWA concentration of 0.62 µg/L being considered the maximum soluble concentration of the test item in test medium. Also the ErC10 was determined to be > 0.62 µg/L. The 72 h NOEC for growth rate inhibition was >= 0.62 µg/L, based on biological relevance (since the measured effect was below 10% of inhibition).
