Description of key information

The LC50 on embryonic stages of zebrafish (Danio rerio) after 96h of exposure to the test item under semi-static conditions was calculated to be 14 mg/L (confidence interval 13.7-14.3 mg/L). The highest tested concentration with no mortality was 4.47 mg/L and the lowest concentration with 100% mortality was 53.4 mg/L.

The acute toxicity of the test item on embryonic stages of zebrafish (Danio rerio) was tested according to the OECD guideline 236 and GLP.

With regard to the test item properties, the study was conducted under semi-static conditions and in the dark. 20 eggs were exposed to the following nominal concentrations of the test item 2.13 - 4.70 - 10.3 - 22.7 - 50 mg/L and to the reference substance. For the control 48 eggs were used including 4 internal control per tested concentration plate. The followig effects coagulation of embryos, absence of somite formation, non-detachment of the tail or lack of heart beat are considered as lethal endpoints which indicate mortality.

The concentrations of the test item were verified by LC-MS/MS at the start of the exposure (0 and 72 h) and at the end of exposure interval (24 and 96h) in all tested concentrations and the control. The measured concentrations were within 102 to 119% of the nominal concentrations at the start of the exposure. At the end of the exposure intervals (24h and 96h) they strongly decreased with the decreasing nominal concentrations resulting in a range of 23% to 105% of nominal values. The geometric mean measured concentrations were calculated to be : 1.43 - 4.47 10.4 - 23.1 - 53.4 mg/L.

Water quality parameters (pH, oxygen saturation and temperature) measured at the start and at the end of exposure were within the acceptable limits. The validity criteria of the OECD Guideline 236 were fulfilled.

The NOEC of the test item was 4.47 mg/L. The LC50 on embryonic stages of zebrafish after 96h of exposure to the test item under semi-static conditions was calculated to be 14 mg/L ( Confidence interval : 13.7 - 14.3 mg/L).
