Acute oral toxicity: 

Acute oral toxicity dose (LD50) of Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) (CAS no: 82338-76-9) was predicted based on OECD QSAR toolbox 5889 mg/kg bw and different studies available on closely related read across substances Terphenyl, hydrogenated (CAS no: 61788-32-7) 17500 mg/kg bw and Benzene, C10-13-alkyl derivs (CAS no: 67774-74-7) > 5000 mg/kg bw. All these studies concluded that the LD50 value is greater than 2000 mg/kg bw. Thus, comparing this value with the criteria of CLP regulation, Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene] cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) can be classified as category V of acute oral toxicity.

Acute Inhalation toxicity: 

Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) (CAS no: 82338-76-9) has very low vapour pressure 2.9E-26 Pa. The normal conditions of use of this substance will not result in aerosols, particles or droplets of an inhalable size, so exposure to humans via the inhalatory route will be highly unlikely and therefore this end point was considered for waiver.

Acute Dermal toxicity dose (LD50) for Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) (CAS no: 82338-76-9) was predicted based on OECD QSAR toolbox 6943 mg/kg bwand differentstudies available for the structurally similar read across substance [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl]methylene] cyclohexa-2,5-dien-1-ylidene]dimethylammonium acetate (CAS No. 83803-79-6) >2000 mg/kg bw and 4, 4’-carbonimidoylbis [N,N-diethylaniline]mono hydrochloride (CAS No. 6358-36-7) >2000 mg/kg bw. All these studies concluded that the LD50 value is>2000 mg/kg bw. Clinical Observations and General Appearance:
Animals were observed for clinical signs, mortality, until sacrifice.
Onset, duration and severity of any sign were recorded. The clinical signs and mortality observations were conducted at 10, 30, 60 minutes, 2, 4 and 6 hours on the day of dosing and once daily thereafter for 14 day. Daily observation was done as far as possible at the same time.
The observations were included general clinical signs, observations of eyes, mucous membranes, respiratory, circulatory system and behavior pattern.

Evaluation of Dermal Reaction:
Dermal reaction was observed daily for study period of 14 days.

Individual animal body weights were recorded pre-test (prior to administration of the test item), day 7 and at termination on day 14.

Gross Pathology:

The study now reported was designed and conducted to determine the acute dermal toxicity profile of 4, 4’-carbonimidoylbis [N,N-diethylaniline]mono hydrochloride (CAS No. 6358-36-7) in Sprague Dawley rats.

The test item was applied to shorn skin of 5 male and 5 female animals at 2000 mg/kg body weight. Administration of the test item at 2000 mg/kg did not result in any skin reaction at the site of application during the study period of 14 days. Administration of the test item did not result in any signs of toxicity and mortality during the study period of 14 days.   

Animals exhibited normal body weight gain through the study period of 14 days.

Gross pathological examination did not reveal any abnormalities attributable to the treatment.

Often are the studies based on in-vivo experiments in rodents, i.e. most commonly in rats for Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) along with the study available on closely related read across substances Terphenyl, hydrogenated (CAS no: 61788-32-7) and Benzene, C10-13-alkyl derivs (CAS no: 67774-74-7). The predicted data using the OECD QSAR toolbox has also been compared with the experimental studies. The studies are summarized as below –

In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, the acute oral toxicity was estimated for Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) (CAS no: 82338-76-9). The LD50 was estimated to be 5889 mg/kg bw, when Wistar male and female rats were treated with Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) via oral gavage route.

The above study is supported by U.S. national Library of Medicine (ChemIDplus, 2017), GESTIS SUBSTANCE Database (2017), and EUROPEAN COMMISSION – European Chemicals Bureau (IUCLID Dataset, 18 – FEB - 2000), for the closely related read across substance Terphenyl, hydrogenated (CAS no: 61788-32-7). Acute oral toxicity study was conducted in 6 Hooded Male rats at the concentration of 17500 mg/kg bw via oral route. 50 % mortality observed at 17500 mg/kg bw. Therefore, LD50 was considered to be 17500 mg/kg bw, when hooded rats was treated with Terphenyl, hydrogenated via oral route.

This study is further supported by RTECS (registry of toxic effect of chemical substance data base, 2017) and EUROPEAN COMMISSION - European Chemicals Bureau (IUCLID Dataset, 19 - FEB - 2000), for the closely related read across substance Benzene, C10-13-alkyl derivs (CAS no: 67774-74-7).Acute oral toxicity study was conducted in rats at the concentration of 5000 mg/kg bw via oral route. No mortality observed at 5000 mg/kg bw. Therefore, LD50 was considered to be > 5000 mg/kg bw, when rats was treated with Benzene, C10-13-alkyl derivs via oral route.

Thus, based on the above studies on Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) (CAS no: 82338-76-9) and it’s read across substances, it can be concluded that LD50 value is greater than 2000 mg/kg bw. Thus, comparing this value with the criteria of CLP regulation, Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) can be classified as category V of acute oral toxicity.

Acute Inhalation toxicity: 

Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) (CAS no: 82338-76-9) has very low vapour pressure 2.9E-26 Pa. The normal conditions of use of this substance will not result in aerosols, particles or droplets of an inhalable size, so exposure to humans via the inhalatory route will be highly unlikely and therefore this end point was considered for waiver.

This study is supported by Sustainability Support Services (Europe) AB (study no.19155, 2017) was designed and conducted for the structurally similar read across substance [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl]methylene] cyclohexa-2,5-dien-1-ylidene]dimethylammonium acetate (CAS No. 83803-79-6). The acute dermal toxicity was tested in Sprague Dawley rats. The test item was applied to shorn skin of 5 male and 5 female animals at 2000 mg/kg body weight. Administration of the test item at 2000 mg/kg did not result in any skin reaction at the site of application during the study period of 14 days. Administration of the test item did not result in any signs of toxicity and mortality during the study period of 14 days. Animals exhibited normal body weight gain through the study period of 14 days. Gross pathological examination did not reveal any abnormalities attributable to the treatment. It was concluded that the acute dermal median lethal dose (LD50) of [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl] methylene]cyclohexa-2,5-dien-1-ylidene] dimethylammonium acetate (CAS No. 83803-79-6), when administered to male and female Sprague Dawley rats was found to be >2000 mg/kg bw. Thus, according to CLP criteria for acute toxicity rating for the chemicals, it infers that [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl] methylene]cyclohexa-2,5-dien-1-ylidene] dimethylammonium acetate (CAS No. 83803-79-6) does not classify as an acute dermal toxicant. CLP Classification: “Unclassified”.

Based on the above studies and prediction on Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) (CAS no: 82338-76-9) and it’s read across substances, it can be concluded that LD50 value is >2000 mg/kg bw for acute oral and dermal toxicity. Thus, comparing this value with the criteria of CLP regulation, Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) can be classified as category V for acute oral and dermal toxicity.For Acute Inhalation toxicity wavier was added so, not possible to classify.
