Justification:

The available evidence on degradation, bioaccumulation potential, and toxicity for C.I. Solvent Blue 98 (3 Amine) indicates that the screening criteria for persistence (P) is met, the screening criteria for bioaccumulation potential (B) is not met, and the screening criteria for toxicity (T) is not met. Based on the information described below, the substance is not PBT nor vP/vB.

Persistence assessment:Each of the evaluated components of C.I. Solvent Blue 98 (3 Amine) were estimated to be not readily biodegradable based on the Biowin outputs (BIOWIN, v4.10). These estimations are deemed reliable because the estimated biodegradabilities of structural analogues agree well with existing experimental data. Based on screening level criteria, the test substance is considered persistent (P).

Bioaccumulation assessment:The predicted log (base 10) of the n-octanol/water partition coefficient forthe components of C.I. Solvent Blue 98 (3 Amine) ranges from 2.89 to 10.98. The empirical determination of the log Kow (based on the OECD 117 HPLC method) indicates a log Kow for the test substance (which is a multi-component substance) ranging from 6.01 to > 6.5. The bioaccumulation potential of C.I. Solvent Blue 98 (3 Amine) was assessed via an EpiSuite QSAR, and the predicted BCF (BCFBAF V3.01) for all of the evaluated components ranged from 9.2to 76.9 L/kg wet weight(i.e.Log BCF = 0.96 to 1.89).  This indicates a low probability of the test material to bioconcentrate in fish. Furthermore,Based on the metabolism prediction by ADMET predictor, allcomponents will be metabolized to hydroxylated metabolites by human CYP 1A2 or CYP 3A4 or combination of human CYP 1A2 and CYP 3A4. The formed aldehyde will be further metabolized to the corresponding acid by aldehyde dehydrogenase (ALDH). The formed metabolites can also be further metabolized to water soluble metabolites (such as glucuronides and sulfates), which will be mainly excreted into urine and feces.Therefore on the basis of low volume of distribution as exemplified by the high empirical Log Kow values, and predicted metabolism and excretion of all components, it can be predicted that this substance is not likely to bioaccumulate. Thus, the test material is not expected to be bioaccumulative (B).

 

Toxicity assessment:The screening assessment for environmental toxicity is based on acute aquatic toxicity testing. No aquatic toxicity testing is available for C.I. Solvent Blue 98 (3 Amine) since the main components are highly water insoluble. To estimate acute aquatic toxicity, EPISUITE ECOSAR (v1.00) was conducted for the main components of C.I. Solvent Blue 98 (3 Amine). ECOSAR estimates indicated that acute toxicity is not expected at concentrations below the predicted water solubility limit for this material, with one minor exception where one modeled component indicated acute toxicity (i.e.96-hr EC50) to algae at a concentration of 26.5 mg/L. Furthermore, reproductive, genotoxicity and repeat dose toxicity studies exposing rats to C.I. Solvent Blue 98 (3 Amine) indicated no concerns for reproductive, mutagenic, or repeat dose toxicity, respectively. Thus C.I. Solvent Blue 98 (3 Amine) does not meet the criteria to be considered toxic (T).
