Reference
Endpoint:
particle size distribution (granulometry)
Type of information:
experimental study
Adequacy of study:
key study
Study period:
2014
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
guideline study without detailed documentation
Qualifier:
according to guideline
Guideline:
OECD Guideline 110 (Particle Size Distribution / Fibre Length and Diameter Distributions) - Method B: Fibre Length and Diameter Distributions
Deviations:
no
Principles of method if other than guideline:
In consideration of the relevance of granulometry study on toxicological assessment, only the size distributions of the sieved fraction < 100 µm were studied in detail.
GLP compliance:
no
Other quality assurance:
ISO/IEC 17025 (General requirements for the competence of testing and calibration laboratories)
Type of method:
microscopic examination
Type of distribution:
counted distribution
Mass median aerodynamic diameter:
9.07 µm
Percentile:
D50
Mean:
9.07 µm
Remarks on result:
other: Migrated from fields under 'Mass median diameter' as D50 percentile. No source field for Standard deviation.

The sample was first sieved by a 100 µm sieve.

weighted sample: 0.5847 g

weight fraction < 100 µm: 0.0066 g

mass fraction < 100 µm: 1.13 %

The mass fraction < 100 µm was 1.13 %, and was investigated by scanning eletron microscopy. The median acpect ration was 2.00 with 0.93 as standard deviation.

The median diameter of the particle size is given for the fraction < 100 µm. The value of 9.07 µm was calculated for spherical particle and a value of 8.19 µm was calculated for cubical particle.

Particle size  Spherical Volume [%]  Cubical Volume [%]
 < 4 µm (respirable)  0.40  0.88
 4 - 10 µm (thoracic)  11.33  21.11
 10 - 100 µm (inhalable)  88.27  78.01
 > 100 µm  0.00  0.00
 Median (diameter µm)  9.07  8.18
Conclusions:
The MMAD (Mass Median Aerodynamic Diameter < 100 μm) has a value of 9.07 μm as spherical particles.
Executive summary:

According to OECD guideline 110, the particle size distribution of 14H-benz[4,5]isoquino[2,1-a]perimidin-14-one was determined by scanning microscopy method for the sieved fraction (< 100 µm, 1.13 % of total sample) and presented on the basis of calculated mass fractions. The median diameter was 9.07 µm with the mean fraction of 88.27 % distribution in the range of 10 to 100 µm; and hence the tested 14H-benz[4,5]isoquino[2,1-a]perimidin-14-one particles with sieved size smaller than 100 µm are mainly dominated by inhalable fraction as defined in EN 481.
