PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
The test substance is a mixture of sparingly water soluble
components. Therefore test solutions were prepared separately
for each test concentration as a water accommodated fraction
(WAF) using a liquid-liquid saturator technique [Ref. 7, 9]. For each
test solution, a glass aspirator bottle (2-L) with a bottom side-outlet
attached to a stopcock is filled with 2-L test media. Then 231-μL
(considering density = 0.8640 g/cm³) of the test substance was
pipetted carefully on the water surface. The bottle was closed and
the solution was stirred on a magnetic plate for about 2 days. The
remaining undissolved test substance remained on the surface
and the required volume of the WAF (approx. 1-L per test solution)
was removed from the bottom port. Undissolved test substance
was visible at the surface of the test aspirator bottle (oily drops).
The control was treated in the same way.
Fresh test solutions were prepared daily. The test vessels were
conditioned with the test solution for 24-h prior to initiating
exposure.

used was supplied by the Institut National de Recherche Chimique Appliquée, France, in 1978. From this date on this clone was cultured and bred continuously in the Laboratory of Ecotoxicology of the Experimental Toxicology and Ecology of BASF SE in Ludwigshafen.
