Various studieshas been investigated for the test chemical3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carbonyl chloride (CAS No: 52314-67-7)to observe the potential for dermal irritation to a greater or lesser extent. The studies are based on in vivo experiments in rabbits for target chemical3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carbonyl chloride (CAS No: 52314-67-7) and its structurally similar read across substancesDicyclopentadiene (CAS no: 77-73-6)and Neodecanoyl chloride (CAS no: 40292-82-8).The predicted data using the OECD QSAR toolbox andDanish QSAR databasehas also been compared with the experimental data and summarized as below;

 

 

In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, the skin irritation potential was estimated for test chemical3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carbonyl chloride (CAS No: 52314-67-7).The chemical 3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carbonyl chloride (CAS No: 52314-67-7) is estimated to be irritating and corrosive to skin of New Zealand White rabbits.

 

 

According toDanish QSAR database, theskin irritation effects were estimated by using four different models i.e, Battery, Leadscope, SciQSAR and CASE Ultra for 3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carbonyl chloride (CAS No: 52314-67-7). Based on estimation, severe skin irritation effects were known when 3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carbonyl chloride (CAS No: 52314-67-7) was exposed to rabbit skin.

 

 

In NTRL report (2008),skin irritation study of structurally similar read across substance Dicyclopentadiene (CAS no: 77-73-6) was conducted on three New Zealand White rabbits to observe its irritation efficacy in accordance with OECD Guidelines No. 404 -Acute Dermal irritation /Corrosion which supports the above result.In this study, 0.5ml of Dicyclopentadienewas applied to intact skin locations on the backs of each rabbits for four hours semi-occlusive condition and skin reactions were scored for 7 daysaccording to Draize scoring system. The results were also interpreted according to the E.E.C. Directive of 29 July 1983 83/467/EEC adapting Council Directive 67/548/EEC on the regulations relating to the classification, packaging and labelling of dangerous substances. Well-defined erythema was observed within 3 days of exposure in all animals. Signs of keratinization were observed on day 7. Moderate edema was observed at 24 hours in all animals, and regressed to slight by day 3. No corrosive effects were observed. Since the Primary irritation index of 4.7was obtained,the chemical Dicyclopentadienewas considered to bemoderately irritatingto the skin ofNew Zealand Whiterabbits .

 

 

The above results were further supported by experimental study conducted by OECD SIDS (2008) ofstructurally similar read across substanceNeodecanoyl chloride (CAS no: 40292-82-8) on four rabbits in order to examine the degree of skin irritation caused by the chemical according to OECD Guideline 404 (Acute Dermal Irritancy/Corrosivity Study). About 0.5 ml of the neat test substance was applied to the intact dorsal skin, clipped 24 hrs before treatment, and left in place under an occlusive cover for 1 hour (2 animals) or 4 hours (2 animals). At the end of the exposure period excess Substance was removed and the skin was rinsed and dried. The Skin reactions were examined and scored immediately after patch removal, and 1, 2, and 8 days thereafter. The skin reactions following 1-hr exposure shown below were reversible within 8 days. Necroses developed in both animals after a 4-hr contact period Full skin necrosis was confirmed by pathological examination of the skin after terminal sacrifice.On the basis of observed effects, the chemicalNeodecanoyl chloride (CAS no: 40292-82-8) can be considered as corrosive and irritatingto the skin of rabbits.

 

 

Thus on the basis of available data for thetarget chemical3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carbonyl chloride (CAS No: 52314-67-7) and its structurally similar read across substancesDicyclopentadiene (CAS no: 77-73-6)and Neodecanoyl chloride (CAS no: 40292-82-8),it can be concluded thatchemical 3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carbonyl chloride (CAS No: 52314-67-7) is able to cause skin irritation and corrosion and thus considered as irritating.Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Category 2 (irritant)”.

The NTRL (1986) and European Communities (2004) reported washed and unwashed primary eye irritation studies ofstructurally similar read across substanceCyclohexane (CAS No: 110-82-7) performed in six male and female rabbits to assess its eye irritation potential. The chemical was installed into the one of each animal and observations were made 24 hours after treatment. In the unwashed study, at one hour post instillation, corneal opacity, involving up to 25% of the cornea, was noted in one rabbit and iritis was noted in another rabbit. Conjunctival redness was noted in five rabbits with conjunctival chemosis in one rabbit. All ocular lesions had cleared within 24 hours and no conjunctival discharge was noted in any of the six animals. In the washed study, Conjunctival redness (vessels injected above normal) was noted in four rabbits at one hour postinstillation and had cleared by 24 hours. No corneal opacity, iritis, conjunctival chemosis, or discharge 'was noted in any of the six rabbits. Since all the observed effects were cleared within 24 hours, the chemical Cyclohexane (CAS No: 110-82-7) was considered to be non-irritant to the eyes of rabbits.

 

 

 

The above results were further supported by an eye irritation study conducted by OECD SIDS (2008) ofstructurally similar read across substanceNeodecanoyl chloride (CAS no: 40292-82-8) on three rabbits in order to examine the degree of eye irritation caused by the chemical according to Draize method. About 1ml of the neat test substance was appliedinto the conjunctival sac of the rabbit'seye.Eye reactions were examined 24, 48, and 72 hrs after application.Reactions of cornea, iris, and conjunctivae were scored according to the table of Draize.No corneal or iridial reaction was noted in any of the animals at any of the examination intervals.  Slight conjunctival reddening was noted in all rabbits 24 hrs and in one animal 48 hrs after treatment. Changes were reversible and absent on day 3 post treatment.A primary irritation index of 1.3 was calculated for test chemical Neodecanoyl chloride (CAS no: 40292-82-8) and thus can be considered as not irritatingto the eye of rabbits.
