In a modified Ames test with vapour exposure of the test substance (supporting study) phosgene revealed no mutagenic activity with the Salmonella typhimurium strains TA 98 and TA 100 in the absence and in the presence of a metabolic activation system (Reichert et al., 1983). As explained by the authors phosgene would not be mutagenic under the conditions of liquid suspension in the Ames test, because this volatile gas reacts rapidly and completely with the components of the incubation system. Unchanged phosgene in the solution was only detected above a gaseous phosgene concentration of 10,000 ppm.

Therefore, further genetic toxicity testing is technically not feasible due to the extreme reactivity and toxicity of phosgene, which is a volatile gas at room temperature and rapidly hydrolyses to carbon dioxide and hydrochloric acid.
